再生障碍性贫血患者骨髓间充质干细胞体外生物学特性与其免疫调节缺陷

背景:许多报道指出骨髓间充质干细胞具有免疫调节及造血功能作用,而再生障碍性贫血患者骨髓间充质干细胞与正常人骨髓间充质干细胞在免疫调节及造血功能都存在显著差异.目的:对再生障碍性贫血患者骨髓间充质干细胞的体外生物学特点及与免疫调节缺陷之间的关系进行综述.方法:由第一作检索Pubmed数据库(1987/2009)和维普数据库(1989/2009),英文检索词为"Mesenchymal stem cell,Bone marrow";中文检索词为"再生障碍性贫血,间充质干细胞",总计检索到文献55篇.纳入与再生障碍性贫血患者骨髓间充质干细胞生物学特性相关及与再生障碍性贫血患者免疫调节相关的文献.排除重复性研究及与主题非密切相关的文献.符合纳入标准的32篇文献中,英文综述3篇,英文研究论文23篇,中文研究论文6篇.结果与结论:骨髓间充质干细胞具有高度增殖、自我更新及多向分化的能力,除支持造血、促进植入及体内造血重建等作用之外,还具有免疫负调节及降低移植物抗宿主病发生率的作用.再生障碍性贫血发病机制与造血干细胞内在增殖或分化缺陷,造血微环境系统异常以及机体免疫功能紊乱等有关,而再生障碍性贫血患者骨髓间充质干细胞的异常可能在其发病机制中存在重要作用.
