金利胶囊对膝骨关节炎兔骨骼肌收缩蛋白基因与软骨基质蛋白多糖表达的影响

摘    要:目的]通过观察免膝骨关节炎(KOA)骨骼肌蛋白与软骨基质蛋白多糖(PGs)表达,探讨柔肝中药金利胶囊治疗KOA的作用及机理.方法]42只新西兰大白兔分为空白对照组(K组),模型对照组(MD组),金利干预组(R组);MD组与R组动物以改良Hulth法造模,R组造模并给药;取膝关节行番红/固绿染色、Mankin评分;RT-PCR检测肌肉α-肌动蛋白(a-actin)、重链肌球蛋白亚型(MHC Ⅰ,MHCⅡa,MHCⅡd/x,MHCⅡb)、Aggrecan与ADAMTS5 mRNA表达,检测关节软骨糖胺聚糖(GAG)水平.结果]与模型组比较,R组Mankin评分降低,α-actin与多种MHC亚型mRNA表达水平上调(P<0.05);Aggrecan mRNA呈高表达,ADAMTS5 mRNA低表达(P<0.05).结论]金利胶囊能够延缓OA软骨退变,促进骨骼肌收缩蛋白基因表达,改善软骨基质蛋白多糖代谢.

Abstract:Objective]To investigate the mRNA expression of α-actin,MHC(myosin heavy chain),ADAMTS5 and Aggrecan in rabbit OA model.To Explore the mechanism of KingLi(liver-softening formulation) in the treatment of knee osteoarthritis.Methods]42 healthy female adult New Zealand white rabbits were randomly divided into three groups(normal group,model group,KingLi group),establishing right knee OA model referring to Hulth’s operation.One week after operation,interfering measures were taken into practice and lasted for 4 weeks.Cartilage sections were made and stained with Safranin’O/Fast Green for Mankin score.The mRNA expression level of α-actin,MHC isoforms in quadriceps femoris,Aggrecan and ADAMTS5 in cartilage was tested by RT-PCR.Results]As to MHC isoforms,the level of MHCⅡa,MHCⅡd/x mRNA in model group was lower than that in normal control group(P<0.05).KingLi was capable of up-regulating the mRNA level of α-actin and two MHC isoforms.The level of Aggrecan mRNA in model group was lower than that in normal control group(P<0.05).The level of ADAMTS5 mRNA in model group was higher than that in normal control group(P<0.05).KingLi was capable of up-regulating the level of Aggrecan mRNA expression(P<0.05)and down-regulating the level of ADAMTS5 mRNA expression(P<0.05).Conclusion] KingLi was capable up-regulating the level of α-actin and MHC mRNA expression.KingLi was capable of delaying degeneration of cartilage in OA animals.KingLi was able to induce the expression of Aggrecan mRNA and inhibit the expression of ADAMTS5 mRNA.
