HPLC检测火腿肠中大豆分离蛋白的方法研究

摘    要:利用高效液相色谱(HPLC)法测定火腿肠中大豆分离蛋白的含量。色谱条件为:Xb ridgeBEH300 C4色谱柱;蒸发光散射检测器;梯度洗脱;流动相A:0.05%三氟乙酸-HPLC级水溶液,B:0.05%三氟乙酸四氢呋喃溶液;流速1.0 mL/min;检测波长254 nm;柱温30℃;进样量20μL。大豆分离蛋白在1%~9%含量范围内与特征峰面积呈良好的线性关系,回归方程为Y=2 923.1 X-970.85,相关系数R=0.994 2,平均回收率为99.30%,RSD为2.04%。HPLC法准确度、精密度、稳定性、重现性良好,为火腿肠中大豆分离蛋白的定量检测提供了可选择的方法。

Abstract:A method for the determination of soy protein isolate in sausage by high performance liquid chromatography(HPLC) was established.The optimum chromatograph parameters were set as following: analytical column as Xb ridge BEH300 C4,evaporative light-scattering detector(ELSD) and gradient elution;mobile phases:A,0.05%TFA in water;B,0.05%TFA in THF;flow rate of mobile phase 1.0 mL/min,detector wavelength at 254 nm,column temperature 30℃ and injection volume 20μL.The linear range of SPI in sausage was 1%-9%.The regression equation was Y=2 923.1X-970.85,and the correlation coefficient was 0.994 2.The average recovery of the method is 99.30%,and RSD is 2.04%.HPLC method is accurate with good precision stability and repetition.It provides an optional analysis for determination of soy protein isolate in sausage and can be used to control the product qualities.
