链霉素、庆大霉素和卡那霉素对豚鼠单个离体毛细胞活性影响的比较

摘    要:为比较氨基甙类抗生素对豚鼠单个离体外毛细胞和前庭毛细胞的影响, 用酶解加机械分离方法获得单个离体的外毛细胞和前庭毛细胞,置于分别含有1 m m ol/L的链霉素、庆大霉素和卡那霉素的培养液中短期培养,并以FDA-PI双染色法鉴定细胞活性。结果发现在仅含上述药物的培养液中, 细胞活性无明显改变, 而在无钙、镁培养液中3 种药物均可引起细胞形态和活性的改变,其中链霉素和庆大霉素对前庭毛细胞活性影响较大,卡那霉素对外毛细胞活性影响较强。临床上链霉素可首选用于经外淋巴灌流治疗梅尼埃病

Abstract:To com pare the differenttoxicities ofam inoglycosides antibiotics on isolated outer hair cell (OHC) and vestibular hair cells (VHC), OHCs and VHCs w ere isolated from guinea pigs by using collagenase digestion and m echanicaldissection. These isolated internalear hair cells w ere subjected to a short-term culture in the m edia containing 1 m m ol/L streptom ycin, gentam icin, kanam ycin HBHBSS respectively, and their viability w as identified by using fluorescein diacetate and propidium iodide double staining procedure. The results show ed thatthe viability ofthe isolated internalear hair cellin the m edia containing the above antibiotics had no obvious change, butthe cellviability and m orphology in free Ca2 / Mg2 preparations w ere affected by gentam icin, streptom ycin and kanam ycin. Streptom ycin and gentam icin had m ore otoxic on VHC than kanam ycin, w hile kanam ycin had m ore otoxic on OHC than streptom ycin and gentam icin. These im plied that streptom ycin w as the m edicine of choice to treat Meriere's disease by perilym phatic perfusion.
