摘    要:目的:观察microRNA分子miR-526 b对非小细胞肺癌细胞A549的化疗敏感性的影响。方法将miR-526 b基因转染到A549细胞中;通过细胞活性检测测定A549细胞对顺铂( diamminedichloroplatinum, DDP)的敏感性,通过流式细胞技术以及Western印迹检测细胞凋亡水平。结果筛选获得稳定表达miR-526 b的克隆细胞。细胞活性检测显示miR-526 b可明显提高非小细胞肺癌细胞A549对顺铂的敏感性,流式细胞技术显示DDP处理后miR-526 b稳定转染细胞的凋亡细胞率(10.2%±1.1%)较对照组A549细胞(3.5%±0.3%)以及DDP未处理对照A549细胞(0.4%±0.1%)明显升高。 Western印迹实验表明DDP处理后miR-526 b非小细胞肺癌细胞A549凋亡指标cleavaged caspase-3、 Bax明显升高。结论 MiR-526 b可促进顺铂诱导非小细胞肺癌凋亡。

Effect of miR-526b Up-regulation on Diamminedichloroplatinum-in-duced Cell Apoptosis in Non-small Cell Lung Carcinoma
Abstract:Objective To investigate the effect of miR-526b on the chemosensitivity of non-small cell carcinoma ( NSCLC) cells A549.Methods Mir-526b gene was transfected into A549 cells.The cell viability assay was used to detect the sensitivity of transfected cells to diamminedichlo-roplatinum ( DDP) , and flow cytometry and Western blotting to detect the cell apoptosis.Results The clone cells stably expressing miR-526 b were screened out.The cell viability assay showed that miR-526b up-regulation could increase the sensitivity of A549 cells to DDP.The flow cytometry showed that the apoptosis rate was 10.2 %±1.1% in A549 cells transfected with miR-526 b and treated with DDP, significantly higher than that in control A549 cells (3.5%±0.3%) and A549 cells given no treatment of DDP (0.4%±0.1%) .Western blotting showed that the apoptotic in-dicators cleavaged caspase-3 and Bax were greatly increased in A549 cells transfected with miR-526b and treated with DDP.Conclusion miR-526b can promote DDP-induced apoptosis in NSCLC.
