维药爱维心口服液对大鼠缺血再灌注心肌ATP敏感性钾通道的影响

摘    要:目的探讨爱维心口服液对大鼠心肌缺血再灌注损伤(myocardial ischemia reperfusion injury,MIRI)的保护作用及机制。方法 40只体质量200~250 g雄性Wistar大鼠,随机分为假手术组,缺血再灌(ischemiareperfusion injury,I/R)组,爱维心预处理组,爱维心预处理+格列苯脲(KATP通道的阻断剂)组四组,各10只。建立大鼠MIRI的模型,记录大鼠心功能指标;检测心肌丙二醛(malondial dehyde,MDA)含量、谷胱甘肽过氧化物酶(activity of glutathione,GSH-PX)活性和血清肌酸激酶(creatine kinase,CK)含量。结果爱维心口服液能够改善大鼠I/R后的心功能,与I/R组比较能显著降低心肌组织MDA和血清CK的含量(P<0.01),GSH-PX的活性明显升高(P<0.01);爱维心预处理+格列苯脲组与I/R组的检测指标基本相似。结论爱维心口服液对大鼠MIRI具有保护作用,其机制可能与抑制膜脂质过氧化,激活ATP敏感性钾通道(KATP通道)有关。

Abstract:Objective To investigate the protective effects and mechanism of Aiweixin on myocardial ischemia reperfusion injury (MIRI) in rats. Methods Forty male Wistar rats weighted 230 ?250 g were divided into four groups randomly: sham operation group, ischemia-reperfusion injury group (I/R) , Aiweixin precondition group, Aiweixin and glibenclamide preconditioning group (n =10 in each group) . The hemodynamic changes and the cardiac muscle contents of malondial dehyde (MDA) , activity of glutathione ( GSH-PX ) and the serum contents of creatine kinase ( CK) were detected respectively, and the therapeutic effects of Aiweixin liquid were observed in rats with MIRI.Results The cardiac muscle contents of MDA and the Serum contents of CK were significantly lower, the activity of GSH-PX were higher in Aiweixin precondition group than that in IR group; there were no differences in these indicators between I/R group and Aiweixin/glibenclamide precondition group. Conclusion Aiweixin liquid can protect MIRI in rats, which may be associated with inhibiting lipid peroxidation of cellular membrane and activating ATP sensitivity of potassium channels.
