| 摘 要： | 目的 探讨人脐血T淋巴细胞活化的可视化特征.方法 将人脐血T细胞在RPM11640完全培养摹中培养,分成两组:实验组加入抗CD3McAb(10 ms/L)和适当浓度的PHA活化;对照组不加CD3McAb和PHA.测定细胞活率,倒置显微镜下观察T细胞活化的特征;制备细胞超薄切片,透射电镜下观察分析并摄影.结果 活化T细胞形态良好,分裂象细胞多数不规则,胞核清晰,体积明显增大;生长旺盛的细胞区域可见到细胞集落.透射电镜下活化T细胞的染色质凝集,核仁数多而明显;典型的活化细胞巾有较多空泡(特征性可视化结构).结论 人脐血T淋巴细胞活化的主要特征是细胞体积增大;染色质凝集,胞质中可出现空泡;形成细胞集落.
