参附芎泽注射液对压力超负荷大鼠血液流动力学和心肌组织及血清TNF-α IL-1β IL-6含量的影响

摘    要:目的:参附芎泽注射液对压力超负荷大鼠血流动力学和心肌组织及血清TNF-α、IL-1β、IL-6含量的影响。方法:参照Ohkusa等介绍的方法复制腹主动脉缩窄大鼠模型。48只SD大鼠随机分为假手术组(12)和造模组(36),术后4周将造模组随机分为模型组(11)、参附芎泽注射液治疗组(11)和依那普利治疗对照组(11)。参附芎泽注射液8mL/(kg.d),依那普利对照组剂量为10mg/(kg.d),假手术组和模型组腹腔注射等剂量生理盐水,干预时间共28天。术后8周,测定血流动学指标后处死动物,用E lASA法测定压力超负荷大鼠血清TNF-α、IL-1β、IL-6含量;用ELASA法测定心肌组织TNF-α、IL-1β、IL-6含量。结果:与假手术组相比,模型组左室内压力峰值(LVSP)、左心室内压最大下降速率(-LVdp/dmt ax)及左心室内压最大上升速率(+LVdp/dtm ax)下降;收缩压(SBP)、舒张压(DBP)、左室舒末期压(LVEDP)均升高;模型组心肌组织及血清TNF-α、IL-1β、IL-6含量均明显高于假手术组(P<0.01)。参附芎泽注射液各项指标均低于模型组(P<0.05和P<0.01),能降低心肌组织及血清TNF-α、IL-1β、IL-6含量,与依那普利组比较无显著性差异(P>0.05)。结论:参附芎泽注射液可改善压力超负荷心衰大鼠血流动力学指标,降低心肌组及血清TNF-α、IL-1β、IL-6含量,阻断这些细胞因子对心肌重塑的影响。

Abstract:Objective:To investigate the effects of ShenFu XiongZe Injection upon the hemodynamics and the levels of TNF-α,IL-1β,IL-6 in the experimental pressure-overloaded rats.Methods:Creating a condition of aorta ventralis coarctation in experimental rats in the way Ohkusa reported. Of the 48 experimental rats,12 are classified randomly to a sham-operation group and 36 to a would-be group,both of which are injected abdominally equal amount physiologic saline and are interfered for 28 day.4 weeks after the operation,the would-be group are further classified randomly to an Injection-treated group(11)in which SheFu QiongZe Injection is applied by 8mL/kg,a model group(11)and a control group treated by 10mL/kg of Enalapril.With the hemodynamics being measured and the popluar being killed 8 weeks later,the levels of TNF-α,IL-1β,IL-6 are measured by RIA method.Results:Comparing with the sham-operation group,in the model group,the LVSP,-LVdp/dtmax and +LVdp/dtmax are lowered,and SBP,DBP,LVEDP are raised.The levels of TNF-α,IL-1β,IL-6 in the would-be group are notably higher than the sham-operation group(P<0.01).In the treated group,all indices are lower than the model group(P<0.05)and(P<0.01),indicating that ShenFu QiongZe Injection could decrease the levels of TNF-α,IL-1βand IL-6,but there is no significant difference comparing with the Enalapril group(P<0.05).Conclusion:ShenFu QiongZe Injection can have an effect on the hemodynamics of the heart failure in the pressure-overloaded rats,that is,the levels of TNF-α,IL-1β and IL-6 are lowered to stop the negative actions from these cytokines.
