肺泡巨噬细胞凋亡对肺间质纤维化的影响

摘    要:目的了解肺泡巨噬细胞的凋亡对肺间质纤维化进展的影响。方法收集一组肺间质纤维化患者的肺泡巨噬细胞(AM),与正常组进行对比,检测两组AM产生细胞因子的活性;然后以干扰素-γ(IFN-1)诱导肺间质纤维化患者AM凋亡,对照诱导前后AM凋亡情况,同时检测诱导前后AM产生细胞因子的活性。结果肺间质纤维化患者AM培养上清液中肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-8(IL-8)水平均明显高于对照组(P〈0.01);AM的诱导凋亡率较自然凋亡率明显增高(P〈0.01);诱导后AM产生细胞因子的活性明显低于诱导前的AM(P均〈0.01)。结论在肺间质纤维化的早期,利用IFN-γ诱导其AM凋亡,就可抑制AM产生细胞因子TNF-α、IL-6、IL-8等,从而控制肺泡炎,阻止肺间质纤维化的进程。

Abstract:Objective To investigate the influence of apoptosis of the alveolar macrophages (AMs) on pulmonary interstitial fibrosis. Methods AMs of the pulmonary interstitial fibrosis patients and healthy controls were collected, the levels of tumor necrosis factor - α ( TNF - α, interleukin - 6 ( IL - 6) and interleukin - 8 ( IL - 8 ) produced by AMs were measured. Then apoptosis of the alveolar macrophages (AMs) were induced by γ-interferon(IFN -γ) and the apoptosis rates of the AMs from pulmonary interstitial fibrosis patients before and after induction was observed and compared, and the levels of TNF - α, IL - 6 and IL - 8 produced by AMs were measured and compared. Results The levels of TNF - α, IL - 6 and IL - 8 produced by AMs of the pulmonary interstitial fibrosis were higher than that of the normal controls ( P 〈 0.01 ). The apoptosis rate of the AMs after induction were significantly higher than that of the natural apoptosis (P 〈 0.01 ), and the levels of TNF - α, IL - 6 and IL - 8 produced by AMs after induction were significantly lower than that produced before induction (P 〈 0. 01 ). Conclusions The induction of apoptisis of AMs by IFN - γin the early stage of pulmonary interstitial fibrosis will reduce the levels of TNF-α IL- 6 and IL- 8 produced by AMs, so as to control the alveolar inflammation and restrain the progress of pulmonary interstitial fibrosis.
