白芥子散对哮喘大鼠免疫稳态重建及RhoA基因表达的影响

摘    要:目的:探讨白芥子散对哮喘大鼠免疫稳态重建及其对于RhoA基因表达的影响。方法:60 只Wistar 大鼠分为六组,A组(正常对照组)、B组(阴性对照组)、C组(哮喘模型组)、D组(地塞米松组)、E组(白芥子散2 h组)以及F组(白芥子散4 h组),每组10只。造模成功后,A组、B组和C组正常饲养; D组腹腔注射地塞米松; E和F组背部剃毛,并在其双侧肺俞、脾俞、肾俞采用白芥子散分别艾灸2 h和4 h。检测和比较六组大鼠支气管肺泡灌洗液中的总细胞数、中性粒细胞数以及巨噬细胞数,白细胞介素-6和TNF-α的表达水平,以及肺组织中RhoA基因的蛋白质表达水平。结果:肺泡组织HE染色结构显示,采用地塞米松和白芥子散治疗2 h和4 h后肺泡结构的破坏,炎性细胞的浸润明显减轻。与正常对照组相比较,哮喘模型组支气管肺泡灌洗液中的总细胞、中性粒细胞和巨噬细胞的数量均显著增加(均P<0.05),TNF-α和IL-6水平均显著增加(均P<0.05),肺组织中的RhoA基因的蛋白质表达水平显著增加。地塞米松和白芥子散2 h和4 h治疗显著减少了支气管肺泡灌洗液中的总细胞、中性粒细胞和巨噬细胞的数量(均P<0.05),TNF-α和IL-6水平均显著增加(均P<0.05),减少了肺组织中的RhoA基因的蛋白质表达水平。结论:白芥子散可改善抗原诱导的大鼠哮喘,助其免疫稳态的重建,其作用机制可能是通过减少RhoA基因的蛋白质表达水平而实现的。

Abstract:Objective:To investigate the effect of Baijiezi powder on immune homeostasis in asthmatic rats and its effect on RhoA gene expression.Methods:Sixty Wistar rats were divided into 6 groups:group A(normal control group),group B(negative control group),group C(asthma model group),group D(dexamethasone group),group E(Baijiezi powder 2 h group )and F group(Baijiezi powder 4 h group),10 mice in each group.After successful modeling,group A,group B and group C were normally reared; group D was intraperitoneally injected with dexamethasone; group E and group F were shaved at the back,and in both sides of Feishu,Pishu and Shenshu were separated by white Baijiezi powder Aijiu 2 h and 4 h.The total number of cells,neutrophils and macrophages,the expression levels of interleukin-6 and TNF-α,and the protein of RhoA gene in lung tissue were detected and compared in the bronchoalveolar lavage fluid of 6 groups of rats.The expression level.Results:HE staining of lung tissue showed that the destruction of alveolar structure and the infiltration of inflammatory cells were significantly alleviated after 2 h and 4 h treatment with dexamethasone and Baijiezi powder.Compared with the normal control group,the total number of cells,neutrophils and macrophages in the bronchoalveolar lavage fluid of the asthma model group were significantly increased(all P<0.05),and the TNF-α and IL-6 levels were significant.The protein expression level of the RhoA gene in lung tissue increased significantly(all P<0.05).2 h and 4 h of dexamethasone and Baijiezi powder significantly reduced the number of total cells,neutrophils and macrophages in bronchoalveolar lavage fluid(all P<0.05),TNF-α and IL-6 The water level increased significantly(all P<0.05).The protein expression level of the RhoA gene in lung tissue.Conclusion: The sputum drug Baijiezi powder can improve antigen-induced asthma in rats and help its immune homeostasis.The mechanism may be achieved by reducing the protein expression level of RhoA gene.
