摘    要:目的:建立一种体外诱导肿瘤细胞特异性γ δ T细胞的培养体系.方法:从人外周血中分离单个核细胞(PBMCs),用不同浓度的磷酸抗原(IPP)与rhIL-2联合刺激γ δ T细胞增殖,观察并收集细胞,采用流式细胞仪检测刺激后 CD3+Vγ9+的 γ δ T 细胞数量以及比例,并计算其扩增效率.结果:经15天扩增培养后,高浓度组10μg/ml 的IPP联合500IU/ml rhIL-2刺激得到的γ δ T细胞的扩增效率最高,且扩增培养至第9天的γ δ T细胞对肿瘤细胞有一定的杀伤活性.结论:适宜浓度磷酸抗原(IPP)联合rhIL-2能有效体外诱导具有较强抗肿瘤活性γ δ T细胞的扩增.

Abstract:Objective:To amply tumor cells specific peripheral blood γδ T from tumor patients.Methods:Isolating peripheral monocytes (PBMCs) from peripheral blood and amplifying γδ T cells by using IPP plus rhIL-2.Analyzing proportion and total cell amount of CD3+Vγ9+ T cells in total PBMCs by using flow cytometry approach.Exploring the tumor cells killing activities of activated CD3+Vγ9+ T cells by analyzing the target cells viability using Cell Counting Kit-8.Results:After 15 days culturing,the group cultured with the medium supplemented with 10μg/ml IPP plus 500IU/ml rhIL-2 showed highest amplifying efficacy.γδ T cells cultured in the 9th day showed optimal tumor cells killing activities.γδ T activated with cell lysates from tumor cells DU145 showed specific cell killing activities.Conclusion:Tumor cells targeting γδ T could be induced and amplified with optimized IPP plus rhIL-2 formula,which could be a potential tool for immune cell therapy.
