二色茉莉组织培养技术体系研究

摘    要:以茎段为材料,采用不同培养基,研究确定适合二色茉莉(Brunfelsialatifolia)组织培养的培养基类型,植株再生的可能途径,以及适宜的培养条件。结果表明:①培养条件为温度25±2℃,光照强度1500~2000lx,光照12h/黑暗12h;②初代培养以MS+BA1 0mg/L+NAA0 01mg/L为最佳组合配方;③接种后直接进行光照培养启动较快;④0 1%升汞对外植体处理时间6min效果最好,启动率达91 5%,污染率2 5%;⑤继代培养以MS+BA2 0mg/L+NAA0 05mg/L为好,其月增殖系数为6 11,丛生芽多且生长适中,30d便可增殖一代;⑥生根培养以1/2MS+IBA2 0mg/L为好,其生根率为93 3%,平均根数为2 85条,平均根长为2 52cm;⑦生根移栽到珍珠岩∶蛭石=1∶1的育苗基质中,30d后成活率为85 9%。

Study on the Technique System of Tissue Culture in Brunfelsia Latifolia
Abstract:In view of studying the suitable type of medium, probable way of explant regeneration and suitable condition of culture for tissue culture in Brunfelsia latifolia, an experimentation was established in which stem segments and a series of different medium were employed. The results are as follows: (1) Culture conditions: temperature 25±2?℃, 12?h illumination every day, and about 1500~2000?lux illumination intensity; (2) Initiation medium: MS+BA 10?mg/L+NAA 001?mg/L is the best one; (3) The directed illumination culture can get good effect; (4) The explants are sterilized in 01% mercuric chloride solution for 6 minutes well. As a result, the rate of initiation is 915% and the rate of pollution is 25%; (5) Subculture medium: MS+BA 20?mg/L+NAA 005?mg/L. The buds grow well. The rate of propagation can reach for 30 days and the increasing coefficient in a month is 611; (6) Rooting medium: 1/2 MS+IBA 20?mg/L, the rooting rate is 933%, the number roots are 285, the length of root is 252; (7) The rooting seedling transplant medium: pearlite∶vermiculite=1∶1, the rate of survive is 859% in 30 days.
