摘    要:目的:采用星点设计-效应面法优选青天葵中总黄酮的提取工艺.方法:采用星点设计进行实验,以乙醇体积分数、乙醇倍量、提取时间为自变量,总黄酮含量为因变量,对各水平进行多元线性回归及二项式拟合,通过效应面法优化工艺参数,并对预测值和实测值进行比较分析.结果:建立的数学模型复相关系数较高,指标与因素的关系适合采用二项式方程拟合(R2 =0.987 0),验证试验结果与模型预测值偏差较小,确定最优提取工艺为30倍量65%乙醇提取2次,每次75 min.结论:该优选工艺简便,可预测性较优,适用于青天葵中总黄酮的开发利用.

Abstract:Objective: To optimize extraction process of total flavonoids from Nervilia fordii by central composite design and response surface methodology. Method: Central composite design was used with the concentration of ethanol,extraction time and the amount of ethanol as independent variables,the content of total flavonoids as dependent variables,each level was done with multiple linear regression and binomial fitting.Technology parameters was optimized by response surface methodology,and predictive analysis was carried out by comparing observed and predicted value. Result: Established mathematical model had high multiple correlation coefficients,relationship between factors and indexes was suitable for fitting with binomial equation(R2=0.987 0), verification test result and model prediction value had small deviation.Optimum extraction process was:extracted 2 times with 30 times the amount of 65% ethanol,75 min per time. Conclusion: This optimized process was simple,and had well prediction,it was suitable for development and utilization of total flavonoids from N. fordii.
