FHIT在结直肠癌中的表达及其与PCNA的关系

目的探讨FHIT与PCNA在结直肠癌中的表达及其与生物学行为的关系,并分析两者的关系。方法应用免疫组化SP法检测FHIT、PCNA在54例结直肠癌及正常结直肠粘膜组织中的表达。结果①FHIT蛋白在结直肠癌中的阳性率为42.59%,在正常组织中的阳性率为88.89%,两者间的差异具有统计学意义（P〈0.01）,FHIT在癌组织的表达明显低于对照组;PCNA蛋白在结直肠癌中的阳性率为75.93%,在正常组织中的阳性率为18.52%,两者间的差异具有统计学意义（P〈0.01）,PCNA在癌组织的表达明显高于对照组;②FHIT在结直肠癌中的表达与淋巴结转移有关（P〈0.01）,FHIT及PCNA与性别、年龄等其他临床病理参数无关;③FHIT与PCNA在结直肠癌中的表达呈负相关（χ^2=6.5074,P〈0.05）。结论FHIT和PCNA的异常表达可能与结直肠癌的发生发展密切相关,两者在肿瘤的恶性进程中扮演重要角色。

Expression of FI-HT protein in colorectal carcinoma and its correlation with the expression of PCNA

Objective To study the relationship between the expressions of FHIT and its bilogical behavior,as well as its correlation with the expression of PCNA in colorectal carcinoma.Methods The expressions of FHIT and PCNA were detected by immunohistochemical staining in 54 case of colorectal carcinoma tissues,and compared with those in 54 cases of normal colorectal tissues.Results ①The positive rate of FHIT protein was 42.59% in colorectal carcinoma tissues and 88.89% in normal colorectal tissues.There existed significant difference between the two groups（P〈0.01）.The positive rate of FHIT protein was lower in tumor than that in normal colorectal tissues.The positive rate of PCNA protein was 75.93% in colorectal carcinma tissues and 18.52% in normal colorectal tissues.There existed significant difference between the two groups（P〈0.01）.The positive rate of PCNA protein was higher in tumorthan that in normal colorectal tissues;②The expressins of FHIT in colorectal carinoma tissues correlates with the metastasis of lymph nodes（P〈0.01）.The expressions of FHIT and PCNA in colorectal carcinoma were not significantly related to the sex of patients or the age of patients and so on（P〉0.05）;③The expressions of FHIT was negatively correlated with PCNA（χ^2=6.5074,P〈0.05）.Conclusion The abnormal expressions of FHIT and PCNA were important genetic events associated with the genesis and development of colorectal carcinoma.
