摘    要:目的观察湿疹1号对血清噬酸性粒细胞阳离子蛋白(ECP)的作用并分析其对T细胞亚群CD4^+、CD8^+及其比值的影响,探讨该药治疗急性脾虚湿疹的作用机制。方法将50只豚鼠随机分成5组,即空白对照组、脾虚急性湿疹空白对照组、脾虚急性湿疹中药治疗低、高剂量组,中药(黄柏胶囊)阳性对照组。采用DNCB涂抹及饮食不节、疲劳过度致脾虚证法建立豚鼠急性脾虚湿疹模型,酶联免疫吸附(ELISA)法与流式细胞术进行血ECP及CD4^+T、CD8^+T的检测。结果与空白对照组比较,模型组血清中ECP过度表达,与模型组比较,各给药组ECP表达均明显下降(P<0.05或P<0.01),且中药高剂量组疗效要优于低剂量和黄柏胶囊组,而低剂量组与黄柏胶囊组的作用差异无统计学意义(P>0.05);与模型组比较,各给药组CD4^+T及CD4^+T/CD8^+T淋巴细胞水平有所升高,而CD8+T水平则有所降低,但仅中药高剂量组差异有统计学意义(P<0.05)。结论湿疹1号能有效抑制皮损中ECP水平的表达,并调节CD4^+T淋巴细胞和CD8^+T淋巴细胞之间的平衡,进而调节人体的免疫功能。

Abstract:Objective In order to observe the effects of Shizhen Yihao on serum macrophage cationic protein (ECP) and than analyze its influence on the CD4+,CD8+ and the ratio of T cells,and to explore the mechanism of the drug in the treatment of acute eczema of spleen-deficiency.Methods Fifty guinea pigs were randomly divided into 5 groups,namely blank control group,acute eczema of spleen deficiency in the blank control group,acute eczema of spleen-deficiency of traditional Chinese medicine in the treatment of low and high-dose group,traditional Chinese medicine (Huangbai capsule) positive control group.Using DNCB smear and diet,fatigue excessive spleen deficiency method to establish the guinea pig models of acute spleendeficiency,enzyme-linked immunosorbent assay (ELISA) and flow cytometry for blood ECP and T CD4+,T CD8+ detection.Results Compared with the blank control group,the serum ECP in model group was over expressed.Compared with the model group,the expression of ECP in each group was significantly decreased (P<0.05 or P<0.01).Effect of the high dose Chinese traditional medicine group was better than that of low dose and Huangbai capsule group,and low dose group and Huangbai capsule group showed no significant difference (P>0.05).Compared with the model group,the drug group,CD4+ T and CD4+/CD8+ T lymphocytes increased,and the level of CD8+ T decreased,but only high dose of Chinese medicine group had significant difference (P<0.05).Conclusion Shizhen Yihao can effectively inhibit the expression of ECP in skin lesions,and regulate the balance between T CD4+ lymphocytes and T CD8+ lymphocytes,and regulate the immune function of the body.
