摘    要:目的     研究酪氨酸激酶Etk/BMX转染对鼻咽癌细胞增殖的影响。方法    利用脂质体2000将野生型酪氨酸激酶Etk/BMX表达质粒pcDNA3.1-Etk导入Etk/BMX低表达水平的鼻咽癌细胞株sune-1中,建立Etk/BMX稳定高表达细胞株sune-wt,空质粒pcDNA3.1转染细胞作为对照(sune-vector)。流式细胞仪检测其转染效率,然后分别进行克隆平板形成率试验和裸鼠成瘤试验。结果    成功筛选到持续高表达Etk的sune-wt细胞,流式细胞仪检测Etk,表达率为(75.5±2.34)%。克隆平板形成率试验sune wt较之sune-1增殖率较高,裸鼠成瘤试验,sune-wt较之sune-vector和sune-1肿瘤生长较快,sune-vector和sune-1肿瘤生长差异无统计学意义。结论    Etk/BMX在鼻咽癌中可促进细胞增殖。

Abstract:Objective    To study the relationship between the tyrosine kinases Etk/BMX and nasopharyngeal carcinoma (NPC) cell proliferation. Methods    Wild type Etk/BMX pcDNA3.1 Etk was recombinated into low expression of Etk/BMX NPC cell line sune-1 using plasmid 2000, and thus the sune-wt cell line which Etk was highly and stably expressed was established, and cells which transfected by pcDNA3.1 empty plasmid (sune-vector) was used as controls.The transfection rate was examined using flow cytometry and then the cells were detected  by a clone plate experiment and inoculated  into nude mice  to observe tumorigenesis. Results    The sune-wt cells with high Etk expression were successfully obtained, showing an expression rate of (75.5±2.34)%. Clone plate experiment showed that grow rate of sune-wt cells was higher than that of sune-vector and sune-1 cells. sune-wt had a higher rate of growth and tumorigenesis than both sune-vector and sune-1 cells in nude mice,but there were no significant differences in tumor growth between sune-vector and sune-1 cells. Conclusion    Etk/BMX may increase the oncogenicity and cell proliferation in nasopharyngeal carcinoma.
