乌司他丁复合氧氟沙星对大鼠肠黏膜二胺氧化酶的影响

摘    要:目的通过检测大鼠肠黏膜中的二胺氧化酶(DAO),来评价乌司他丁复合氧氟沙星对脓毒症大鼠肠黏膜的保护作用。方法脓毒症大鼠80只,随机均分为四组:空白对照组(C组)、氧氟沙星组(O组)、乌司他丁组(U组)和氧氟沙星加乌司他丁组(O+U组),每组再分为12h和24h两组。O组给予氧氟沙星8mg/kg,U组给予乌司他丁15×104U/kg,O+U组则同时给予氧氟沙星8mg/kg和乌司他丁15×104U/kg,2次/d,连续2d,C组给予生理盐水4ml/kg作为对照。在治疗后24h和48h取大鼠肠组织检测DAO,观察肠黏膜DAO的变化情况。结果肠组织DAO在治疗组中均明显升高,O组、U组与C组比较差异有统计学意义(P<0.05),O+U组与C组比较差异有显著统计学意义(P<0.01),O+U组与O组、U组比较差异有统计学意义(P<0.05),O组与U组之间差异无统计学意义(P>0.05)。结论乌司他丁能保护肠黏膜的屏障功能,增加肠黏膜中DAO的活性,与抗感染药物合用效果更为明显。

Abstract:Objective To evaluate the protection of ulinastatin combined with ofloxacin to diamine oxidase (DAO) of intestinal mucosa in pyemic rats.Methods A total of 80 septic rats were randomly divided into four sets equally: control group (Group C), ofloxacin group (Group O), ulinastatin group (Group U) , and ofloxacin and ulinastatin group Group (O+U)], each of which was then divided into two sets (i.e. 12-hour group and 24-hour group). Ofloxacin was used with 8 mg/kg in Group O, ulinastatin was used with 15×104 u in Group U, and ofloxacin was used with 8 mg/kg and ulinastatin was used with 15×104 u at the same time in Group (O+U) twice a day for two days. Normal saline was used with 4 ml/kg in Group C. Rats’ intestinal mucosal samples were detected for DAO respectively after 24 and 48 hours' treatment in order that the variation of DAO would be observed.Results DAO was upgraded obviously in the three trial sets. There was significant difference between Group O or Group U and Group C (P<0.05), and more significant difference between Group (O+U) and Group C (P<0.01). Furthermore, there was significant difference between Group (O+U) and Group U or Group O (P<0.05), and there wasn’t significant difference between Group O and Group U (P>0.05). Conclusion Ulinastatin helps to protect the barrier function of intestinal mucosa, and to increase the activity of DAO in intestinal mucosa. The effect will be better when it is used in combination with anti-infective drugs.
