RP—HPLC测定拉莫三嗪和苯巴比妥的血药浓度

目的：建立RP—HPLC法同时测定人血清中抗癫痫药拉莫三嗪和苯巴比妥浓度的方法。方法：采用AgilentZorbax XDB—C18（4．6mm×150mm，5μm）色谱柱；流动相：0．1％乙酸-乙腈，梯度洗脱；检测波长：240nm；流速：1．0ml／min。结果：拉莫三嗪和苯巴比妥的血药浓度分别在0．02-1．00μg、0．02-3．00μg范围内与峰面积呈良好的线性关系，平均回收率分别为97．62％和97．96％，拉莫三嗪2．0、20．0、40．0μg/ml日内精密度（RSD，n=5）分别为4．3％、3．9％和2．9％，日间精密度（RSD，n=5）分别为5．1％、5.4％％和3．9％；苯巴比妥5．0、20．0和50．0μg/ml日内精密度（RSD，n=5）分别为3．5％、4．2％和3．6％，日间精密度（RSD，n=5）分别为5．0％、4．7％和4．2％。结论：本方法简便、准确、重现性好，可用于临床上常规监测拉莫三嗪和苯巴比妥的血药浓度。

Content determination of Lamotrigine and Phenobarbital in human plasma by RP-HPLC

Objective： To establish a method for the content determination of lamotrigine and phenobarbital in human plasma by RP-HPLC. Methods： The Agilent Zorbax XDB-C18 （4.6 mm×150 mm, 5 μm） column was used. The mobile phases consisted of 0.1% acetic acid and acetonitrile, gradient clution. The flow rate was 1.0 ml/min and the detection wavelength was at 240 nm. Results： The linear ranges of Diphenhydramine Hydrochloride were at 0.02-1.00 μg and 0.02-3.00μg, respectively. Lamotrigine presented a good linear relationship in the range of 1.00-50.0 μg/ml （r=0.999 1）; phenobarbital in the range of 1.00-150.0μg/ml （r=0.999 0）. The intra-day RSD （n=5） of 2.0, 20.0 and 40.0μg/ml of lamotrigine were 4.3%, 3.9% and 2.9%; the inter-day RSD （n=5） were 5.1%, 5.4% and 3.9%. The intra-day RSD of 5.0, 20.0 and 50.0 μg/ml of phenobarbital were 3.5%, 4.2% and 3.6% （n=5）; the inter-day RSD were 5.0%, 4.7% and 4.2% （n=5）,respectively. The recovery rates （n=5） of lamotrigine and phenobarbital were 97.62% and 97.96%, respectively.Conclusion： The method is fast, reliable, accurate and can be used in monitoring plasma levels of lamotrigine and phenobarbital in clinic.
