探索姜黄二酮和月桂酸共同靶基因BDKRB2在前列腺癌中的治疗意义

摘    要:目的基于月桂酸和姜黄二酮的抗肿瘤特性,发掘前列腺癌中新的靶向治疗基因。方法BATMAN-TCM数据库预测2种化合物共同靶蛋白基因,metascape数据库对共同靶点进行功能富集分析确定关键基因,GEPIA数据库筛选研究靶点。对研究靶点进行生存分析、tdROC曲线分析以及GSEA分析。结果月桂酸和姜黄二酮两者共有24个靶蛋白基因,功能富集分析确定8个关键基因和前列腺癌相关,其中BDKRB2基因在肿瘤组织和正常组织之间差异表达。生存分析显示,BDKRB2低表达与患者不良预后相关。单基因GSEA提示,BDKRB2基因通过多种信号通路与前列腺癌发生发展相关。结论月桂酸和姜黄二酮可共同作用于BDKRB2基因,从而在前列腺癌中发挥治疗作用。

Abstract:Objective To explore a novel gene for targeted therapy of prostate cancer based on the anti-tumor properties of lauric acid and curdione.Methods BATMAN-TCM database predicted the common target genes of the two compounds;the metascape database carried out functional enrichment analysis on the common target to determine the key genes;the GEPIA database screened the target.Survival analysis,tdROC curve analysis and GSEA analysis were performed on the target.Results There were 24 target genes of lauric acid and curdione,and functional enrichment analysis identified 8 key genes associated with prostate cancer,among which,BDKRB2 was differentially expressed between tumor and normal tissues.Survival analysis showed that low BDKRB2 expression was associated with poor prognosis.Single gene GSEA suggested that BDKRB2 was associated with the development of prostate cancer through multiple signaling pathways.Conclusion Lauric acid and curdione can act on the BDKRB2 together,thus playing a therapeutic role in prostate cancer.
