大孔吸附树脂纯化核桃内种皮总多酚的工艺优选

摘    要:目的:优选大孔吸附树脂纯化核桃内种皮总多酚的工艺条件。方法:以核桃内种皮为原料,采用乙醇回流法提取总多酚,通过静态吸附-洗脱试验筛选大孔吸附树脂型号,通过单因素试验考察上样液质量浓度、上样量、吸附流速、洗脱液浓度和用量等对核桃内种皮总多酚大孔树脂纯化工艺的影响。结果:选择X-5型大孔树,其最佳纯化工艺条件为上柱液pH 5.0,质量浓度3 g·L-1,吸附流速3 BV·h-1,上样量9.5 BV,加70%乙醇2 BV洗脱,收集洗脱液;总多酚纯度由粗提物的64.52%提高至79.64%,转移率89.83%。结论:优选的纯化工艺稳定可行,X-5型大孔树脂适用于核桃内种皮总多酚的初步纯化。

Abstract:Objective: To optimize purification technology of total polyphenols from Juglandis Semen kernel pellicles by macroporous resin. Method: With Juglandis Semen kernel pellicles as raw material,total polyphenols was extracted by ethanol refluxing method,static adsorption-elution test for total polyphenols were carried out to select proper macroporous resin,single factor tests were adopted to optimize purification process by taking the concentration of sample solution,adsorption flow rate,eluent dosage and so on as factors. Result: X-5 macroporous resin showed the best separation efficiency by comparing with other resins,its optimum purification technology was as follows:the concentration of sample solution 3.0 g·L-1 with pH of 5.0,adsorption rate of 3 BV·h-1,sample volume of 9.5 BV,eluted with 2 BV of 70% ethanol,collected eluent;Purity of total polyphenol was enhanced from 64.52% to 79.64% with transfer rate of 89.83% in extract. Conclusion: This optimized process was stable and feasible,X-5 resin was suitable for initial purification of total polyphenols from Juglandis Semen kernel pellicles.
