| 摘 要： | 目的为了研究益康唑(Econazole)诱导鼠粒单核白血病细胞 WEHI-3凋亡的机制。方法利用 Annexin-V/PI 双染实验测定细胞凋亡,Fura-2荧光负荷技术检测细胞内游离钙离子浓度([Ca~(2+)]i),并抽取 WEHI-3细胞内质网部分蛋白,利用 Western blot 测定 Caspase-7、Caspase-12的蛋白表达。结果 Econazole 作用后 WEHI-3出现典型的凋亡改变。细胞内游离[Ca~(2+)]i 较对照明显升高,Western blot 结果显示随着细胞内钙离子浓度的提高,Caspase-12和 Caspase-7表达逐渐增加。结论 Econazole 能诱导 WEHI-3细胞凋亡,caspase-12在此过程中起重要作用。

| Abstract: | Objective:To investigate apoptosis in mouse leukemia cell (WEHI-3) induced by Econazole and its mechanism.Methods:Apoptosis induced by Econazole was examined by flow cytometry.Free calcium ([Ca~(2+])i) was determined by Fura-2 fluorescein load technique.The protein was isolated from endoplasmic reticulum of WEHI-3 cells,and then the expression of caspase-12 and caspase-7 was detected by Western blot.Results:WEHI-3 cells exhibited typical change of apoptosis when they were treated by Econazole. [Ca~(2+)]i was significantly higher in Eeonazole-treated group than in control group.The expression of caspase- 12 and caspase-7 was increased with the increase of Econazole concentration.Conclusion:Caspase-12 may play a key role in WEHI-3 apoptosis induced by Econazole. |
