含CpG寡脱氧核苷酸对小鼠肝癌的生长抑制作用及造血功能的影响

摘    要:目的 观察含CpG寡脱氧核苷酸 (CpGODN)对种植小鼠肝癌 (hepatoma 2 2 ,H2 2 )的生长抑制作用及对荷瘤小鼠造血功能的影响。方法 右股皮下接种H2 2 细胞后的BALB/C小鼠 ,随机分为 7组 ,包括生理盐水组、非CpGODN组、CpGODN组、5 氟尿嘧啶 (5 FU)组、白细胞介素 2 (IL 2 )组、CpGODN +5 FU组和IL 2 +5 FU组。皮下注射给药 ,给药期间鼠尾静脉采血观察骨髓抑制情况。用药 12天 ,于最后一次用药 2 4h后颈椎脱臼处死小鼠 ,剥离肿瘤 ,测量大小和称重。结果 单用CpGODN、CpGODN +5 FU、5 FU、IL 2 +5 FU的抑瘤率为 4 2 75 %、5 6 0 2 %、6 3 98%、5 7 91% (P <0 0 1) ,后两组不良反应明显 ,有显著骨髓抑制作用。结论 CpGODN对移植小鼠肝癌生长具有抑制作用 ,并能缓解化疗引起的骨髓抑制状态。

Abstract:Purpose To observe the effects of oligodeoxynucleotide containing CpG motifs on inducing hepatoma rejection and hemopoiesis. Methods H 22 cells were transplanted subcutaneously into the right back legs of BALB/C mice to establish tumor. They were divided into seven groups at random, including normal saline(control), non-CpG ODN, CpG ODN, 5-FU, IL-2, CpG+5-FU and IL-2+5-FU, which were injected subcutaneously nearby the tumor. Inhibition of bone marrow during treatment was observed by using blood of the vein of mice tail. After 24 hours of the last injection at the twelfth day, mice were killed, and the tumors were then measured and weighted. Results The rates of tumor inhibition of CpG, CpG+5-FU, 5-FU and IL-2+5-FU groups were 42.75%, 56.02%, 63.98% and 57.91% (P<0.01) respectively. The last two groups had obviously bad side reaction and bone marrow inhibition. Conclusions CpG ODN inhibits growth of the transplantated tumor of H 22 and enhances hemopoietic regeneration from myelosuppression as caused by chemical therapy.
