摘    要:目的:从地稔中提取分离地稔多糖,并对单糖组成进行研究。方法:采用水提醇沉法提取地稔粗多糖,再用DEAE纤维素柱色谱进行分离纯化,获得一种水溶性较好的中性多糖和一种酸性多糖,然后采用高效液相柱前衍生法对其单糖组成进行测定。结果:地稔中性多糖主要是由葡萄糖、半乳糖、甘露糖、阿拉伯糖、鼠李糖等5种单糖组成,摩尔比为123∶84∶27∶19∶5,另有极少量木糖。地稔酸性多糖单糖组成主要是半乳糖、半乳糖醛酸、葡萄糖、阿拉伯糖、甘露糖、葡萄糖醛酸、鼠李糖7种单糖组成,摩尔比为14∶10∶7∶6∶4∶1∶1,另外还有少量的木糖。结论:本研究建立了分离及测定地稔多糖组成成分的分析方法,该方法操作简单,方便可行。

Abstract:Objective: To extract and separate polysaccharide from Melastoma dodecandrum and to analyze the monosaccharide composition. Method: The polysaccharide was extracted by the water and precipitated by alcohol, and then separated and purified by DEAE-cellulose column chromatography. Water-soluble neutral polysaccharide and acidic polysaccharide were obtained. The monosaccharide composition of the purified polysaccharides was determined by HPLC method with precolumn derivatization. Result: The neutral polysaccharide was consisted of glucose, galactose, mannose, arabinose, and rhamnose, with molar ratio of 123:84:27:19:5, also contained a small amount of xylose. The acidic polysaccharide was consisted of galactose, galacturonic acid, glucose, arabinose, mannose, glucuronic acid, and rhamnose, with molar ratio of 14:10:7:6:4:1:1, also contained a small amount of xylose. Conclusion: To develop an analysis method for extracting polysaccharide from M. dodecandrum and determining it's composition, the method is simple, convenient and feasible.
