原花青素降低家兔动脉血压的机制研究

摘    要:目的:研究葡萄籽提取物原花青素(procyanidins,PC)降低家兔动脉血压的作用及机制。方法:采用颈总动脉插管测量动脉血压,将动物随机分为:PC、左旋硝基精氨酸、甲烯蓝、异搏定、普萘洛尔、吲哚美辛、缬沙坦7组。PC组为静脉累积注射PC(4.0、8.0、16、32、64、84mg/kg)后,记录血压并计算下降百分率(%),其余各组先注射不同的阻断剂对血压无影响后。再注射PC(64mg/kg)测量各组血压并计算下降百分率(%).结果:PC能显著降低家兔动脉血压,并具有量效依赖性(收缩压:r=0.92,t=17.44,P〈0.01;舒张压:r=0.92,t=17.47,P〈0.01)。左旋硝基精氨酸和甲烯蓝能明显减弱PC(64mg/kg)的降压作用(P〈0.01);异搏定、普萘洛尔、吲哚美辛、缬沙坦对PC的降压作用无明显影响。结论:PC低家兔动脉血压的机制可能与内皮释放的NO有关。

Abstract:Objective : To study the decreasing arterial blood pressure of the procyanidins (PC) on rabbit in vivo and the possible mechanism. Methods: Rabbit common carotid artery was intubated and arterial blood pressure in vivo was recorded. Rabbits were divided into the control groups, PC groups, PC add blocker groups at random. The control groups: 0.9% sodium chloride was injected into vein. PC groups: PC with cumulative dose of 4.0,8.0, 16,32,64,84mg/kg was injected into vein and the changes of arterial blood pressure were recorded. PC add blocker groups were divided into five groups including PC+L-NNA, PC+MB, PC+Vpm, PC+Prop, PC+Indo, PC+Val. After the blocker injected into vein had no effects on the arterial blood pressure, PC(64 mg/kg) was injected into vein respectively and the changes of arterial blood pressure were observed. Results : PC has a significant dose-effect dependence on arterial blood pressure in vivo (The systolic pressure:r=0.92, t=17.44, P〈0.01; The diastolic pressure: r=0.92, t=17.47, P〈0.01). The effect of the decreasing arterial blood pressure of PC was significantly blocked by L-NNA and MB. Conclusion : PC can decrease arterial blood pressure significantly in vivo. The effect of decreasing arterial blood pressure of PC 见 may relate to NO releasing from Endothelial cells.
