HPLC法测定导赤丸中黄芩苷和大黄素的含量

目的:建立测定导赤丸中2种有效成分黄芩苷和大黄素的含量的方法。方法:采用高效液相色谱法。色谱柱为Chroma-trex C18柱;测定黄芩苷的流动相为甲醇-水(加入磷酸调pH=3.5,43:57),检测波长为280nm,流速为1.0mL·min-1,柱温为30℃;测定大黄素的流动相为甲醇-水(加入冰醋酸调pH=2.0,80:20),检测波长为245nm,流速为1.0mL·min-1,柱温为25℃。结果:黄芩苷和大黄素的检测浓度分别在0.626～1.044mg·mL-1(r=0.9996)和5～25μg·mL-1(r=0.9998)范围内线性关系良好;平均回收率(n=9)分别为100.18%(RSD=0.75%)和99.30%(RSD=1.5%)。结论:本方法操作简便、准确度高、专属性强,适用于导赤丸的质量控制。

Content Determination of Baicalin and Emodin in Daochi Pills by HPLC

OBJECTIVE：To establish the method for the content determination of baicalin and emodin in Daochi pills.METHODS：HPLC was adopted with chromatrex C18 as column.The mobile phase for baicalin determination consisted of methanol-water-phosphoric acid（43：57） with detection wavelength at 280 nm.The flow rate was set at 1.0 mL·min-1 and the column temperature was 30 ℃.The mobile phase for emodin determination was methanol-water-glacial acetic acid（80：20） with detection wavelength at 245 nm.The flow rate was 1.0 mL·min-1 and the column temperature was 25 ℃.RESULTS：There was good linearity between the peak area and concentration within the ranges of 0.626～1.044 mg·mL-1（r=0.999 6） for baicalin and 5～25 μg·mL-1（r=0.999 8） for emodin,respectively.The average recoveries of baicalin and emodin were 100.18%（RSD=0.75%）and 99.30%（RSD=1.5%）,respectively（n=9）.CONCLUSION：The method is simple,accurate and specific for quality control of Daochi pills.
