雌激素减轻大鼠血管钙化的实验研究

摘    要:目的探讨雌性激素在血管钙化中的作用。方法制备30例维生素D3加尼古丁诱导雌性大鼠血管钙化模型,比较去势和去势补充雌激素对血管钙化的影响。放射免疫法测定血浆雌激素水平,并进行VonKossa染色、钙含量、45Ca2+沉积及碱性磷酸酶(alkalinephosphatase,ALP)活性测定。结果钙化组大鼠血管主动脉平滑肌细胞及其间质内有大量黑色颗粒沉积,主动脉钙含量、45Ca2+沉积及ALP活性分别较对照高2.5倍、3.3倍和2.4倍(均P<0.01)。预先去势的动物(去势+钙化组)钙含量、45Ca2+沉积及ALP活性分别较单纯钙化组升高28%、34%和20%(均P<0.01)。补充雌激素减轻去势+钙化组钙含量、45Ca2+及ALP活性分别低37%、37%和31%(均P<0.01)。结论去卵巢大鼠主动脉钙化程度加重,补充雌激素能减轻钙化,提示雌激素具有保护血管、拮抗钙化的效应。

Abstract:Objective To explore the role of Estrogen in vascular calcification.Methods Thirty female SD rats were ovariectomized and induced vascular calcification by Vitamin D3 and Nicotine. The level of Estrogen was measured by radioimmunoassay, and calcium,~45 Ca~2+ deposition and alkaline phosphatase(ALP)were also assayed.Results Many black granules among the smooth muscle cells and its matrix in calcified aorta by Von Kossa staining in VDN group was observed, 2.5-fold, 3.3-fold, 2.4-fold increase Calcium、~45 Ca~2+ deposition and ALP were observed in VDN group than that in control group, respectively. However, compared with VDN group,these data were increased by 28%、34% and 20% in OVX+VDN group and decreased by 37%、37% and 31% in OVX+VDN+E group,respectively.Conclusion The extent of Vascular calcification is aggravated in ovariectomized rats. It suggests that estrogen may have cardiovascular protection by antagonizing vascular calcification.
