| 摘 要： | 为提高间接ELISA法检测抗-HCV诊断试剂盒的灵敏度,将含有十二烷基硫酸钠（SDS）与巯基乙醇（2-ME）的包被液对HCV重组抗原的固相化,与不加入上述上述两种化学物质的包被液进行对比。结果发现,在包被液中加入适宜浓度的SDS和2-ME,有效的提高了间接ELISA检测抗-HCV试剂的灵敏度,而试剂的特异性则保持不变,优化了临床标本的反应模式。在包被液中同时加入SDS和2-ME,有效的改变了HCV重组抗原的包被效果,优化了抗-HCV诊断试剂盒的生产工艺。

| Abstract: | To improve the sensitivity of anti-HCV ELISA kit,the coating buffer solution of HCV recombinant antigen containing SDS and 2-ME were used to coat on the microwell plates.The coated plates were used in anti-HCV ELISA kit for testing and also compare with plates which were coated with normal buffer solution.The results showed the coating buffer solution containing SDS and 2-ME significantly improved the coating efficiency and the sensitivity of antigen-antibody reaction,and the specific activity of agents wer... |
