骨肉瘤-巨噬细胞共培养体系中单核细胞趋化蛋白-1的表达

摘    要:目的 研究骨肉瘤细胞及肿瘤浸润巨噬细胞中单核细胞趋化蛋白 1(MCP 1)的表达情况。方法 建立骨肉瘤细胞与巨噬细胞的transwell体外共培养模型 ,以模拟体内骨肉瘤组织的微环境 ,应用RT PCR法及ELISA法检测 2种细胞中MCP 1表达情况。结果 共培养前 2种细胞中均表达MCP 1,共培养后骨肉瘤细胞中MCP 1的表达水平变化不明显 ,而巨噬细胞中其水平则显著变化 (P <0 .0 1)。结论 在骨肉瘤组织中浸润的巨噬细胞可能是MCP 1的重要来源之一 ,可通过正反馈的机制使更多单核细胞向肿瘤局部组织浸润 ,从而对肿瘤的生物学行为发生影响。

Abstract:Objective To investigate the expression of MCP-1 in osteosarcoma cells and infiltrating macrophages in osteosarcoma tissues.Methods An in vitro osteosarcoma-macrophage coculture transwell model was established to simulate the in vivo conditions.MCP-1 expressions in the two cell lines were measured using RT-PCR and ELISA methods.Results MCP-1 was constitutively expressed in both cell lines before coculture.After coculture,MCP-1 expression was significantly altered in macrophages(P<0.01),but not in osteosarcoma cells.Conclusion Tumor infiltrating macrophages may be an important source for MCP-1.A positive feedback mechanism may result in more infiltrating macrophages and make an influence on the biological behaviors of tumors.
