辛伐他汀对大鼠脑缺血再灌注后脑组织NOS表达的影响

摘    要:目的观察辛伐他汀对大鼠脑缺血再灌注后脑组织eNOS、nNOS和iNOS表达的影响。方法24只Wistar大鼠随机分为假手术组(Sham)、缺血再灌注IR组(IR)、辛伐他汀预处理高、低剂量组,每组6只。高剂量组和低剂量分别以阿伐他汀6mg/kg和1.5mg/kg连续灌胃7天。用线栓法制备大脑中动脉缺血再灌注模型,缺血2h,再灌注22h,应用免疫组化法检测脑组织中eNOS、nNOS和iNOS的表达。结果与缺血再灌注组比较,辛伐他汀低剂量组eNOS表达差异无显著意义(P>0.05),高剂量组eNOS表达显著增加(P>0.05);高剂量和低剂量组nNOS和iNOS的表达显著降低(P<0.01,P<0.05)。结论辛伐他汀预处理能抑制大鼠脑缺血再灌注后脑组织iNOS、nNOS的表达、上调eNOS表达。

Abstract:Objective To observe the effects of simvastatin on expression of NOS in rats with focal cerebral ischemia reperfusion.Methods 24 Wistar rats were randomly divided into four groups:sham group,ischemia-reperfusion group(IR),simvastatin preconditioning in high dose group(H-Sim)and low dose groups(L-Sim).H-Sim and L-Sim were administrated Sim 6mg/kg and 1.5mg/kg by douche via stomach once a day for 7 days.The focal cerebral ischemia model was established in Wistar rats by using the method of thread inserting left middle cerebral artery occlusion.The expression of eNOS,nNOS and iNOS were measured by immunohistochemistry.Results Compared with IR group,the expression of eNOS in L-Sim did not significant difference,but obviously higher in H-Sim,expression of nNOS and iNOS were decreased significantly in H-Sim and L-Sim(P<0.01,P<0.05).Conclusion Simvastatin can downregulate the expression of iNOS and nNOS and upregulate the expression of eNOS in rats with focal cerebral ischemic-reperfusion.
