人腹主动脉瘤与动脉粥样硬化闭塞症基因表达谱差异的研究

摘    要:目的探讨人腹主动脉瘤与动脉粥样硬化闭塞症的基因表达谱,筛选差异表达基因。方法选取腹主动脉瘤、粥样硬化闭塞动脉标本及正常腹主动脉标本各5例。抽提总RNA,转录并以生物素标记后,与芯片杂交,并对结果进行分析。结果腹主动脉瘤与动脉粥样硬化闭塞比较上调基因119个,下调基因98个,动脉粥样硬化闭塞与正常组织比较,上调基因159个,下调基因84个,腹主动脉瘤与正常组织比较上调基因283个,下调基因310个。而且发现与炎症发应、免疫反应及某些化学趋化因子有关的基因在腹主动脉瘤和动脉粥样硬化闭塞中均上调,腹主动脉瘤与动脉粥样硬化闭塞比较,与自身免疫反应有关的基因明显上调,与钙离子通道调节蛋白及细胞骨架有关的基因明显下调。结论表达谱芯片筛选腹主动脉瘤与动脉粥样硬化闭塞的差异基因,为研究腹主动脉瘤和动脉粥样硬化闭塞的发病机制提供新思路,多种基因可能在动脉粥样硬化向腹主动脉瘤和动脉粥样硬化闭塞的发展过程中起着重要的作用。

Abstract:Objective To screen the differently expressed genes in AAA and ASO by microarray. Methods Five abdominal aorta aneurysm specimens, 5 atherosclerosis specimens and 5 normal abdominal aorta specimens were collected respectively and total RNA was extracted and purified from the samples. Then, labeled cDNA was synthesized and hybridized with Affymetrix GeneChip. Finally, the data a-nalysis was performed to select differentially expressed genes. Results 119 genes were up -regulated in AAA comparing with ASO, 98 genes downregulated; In ASO 159 genes were up regulated when compared with normal artery, 84 genes downregulated; In AAA, 283 genes were upregulated comparing with normal artery, 310 genes downregulated. The genes involved in the functions of immunoreaction, inflammation and chemokine were upregulated in both AAA and ASO. Compared with ASO, the genes involved in the functions of immune response were upregulated, and those involved in the functions of calcium ion transport and structural constituent of cytoskeleton were downregulated. Conclusion The investigation based on microarray in researching differently expressed genes in AAA and ASO affords a new idea in studying pathogenesis of AAA and ASO. Many genes may play an important role in the process of arteriosclerosis to AAA and ASO.
