博莱霉素致大鼠肺纤维化形成过程中Thy-1表达变化与细胞因子生成之间关系的研究

摘    要:目的 测定博莱霉素致肺纤维化动物模型支气管肺泡灌洗液(bronchoalveolar lavage fluid,BALF)中白介素1(interleukin-1,IL-1)、肿瘤坏死因子α(tumor necrosis factor-α,TNF-α),研究其与肺成纤维细胞表面Thy-1表达阳性率之间的关系,探讨肺纤维化发病机制.方法 发于不同时间点采用免疫组织化学法测定Thy-1表达阳性率,采用胸腺细胞法测定BALF中IL-1的活性,采用结晶紫法测定TNF-α活性.结果 肺纤维化形成过程中,成纤维细胞表面Thy-1表达阳性率呈进行性下降,与IL-1、TNF-α活性之间无相关性.结论 经 肺纤维化形成与Thy-1表达之间密切相关,Thy-1减少与IL-1、TNF-α无明显相关性,可能是一个综合因素作用的结果 .

Abstract:Objective To measure expression of Thy-1 and content of cytokine in bronchoalveolarlavage fluid(BALF) in animal models of pulmonary fibrosis induced by bleomycin and observe relationshipbetween expression of Thy-1 and cytokine and to approach pathogenesis of pulmonary fibrosis. MethodsExpression of Thy-1 was observed by immunohistochemistry. Activity of interleukin-1 (IL-1) and tumornecrosis factor-α(TNF-α) in BALF were measured by thymocyte method and genitian violet respectively.Relationship between expression of Thy-1 and cytokine was analyzed. Results Expression of Thy-1 infibroblasts decreased. Expression of Thy-1 about pulmonary fibrosis was uneorrelated with IL-1 and TNF-α.Conclusions Pulmonary fibrosis is closely correlated with Thy-1 expression. There are no obvious relevancebetween reduction of Thy-1 expression and IL-1 ,TNF-α. The disease may be a result of complex factors
