摘    要:目的:探讨免疫电镜胶体金技术在应用中某些关键技术环节的处理。方法:应用免疫电镜胶体金技术对大肠癌GST-Pi(胎盘型谷胱苷肽S转移酶)和金黄地鼠胚胎神经管NGF(神经生长因子)和bFGF(碱性成纤维细胞因子)进行定位观察。结果:大肠粘膜细胞和神经上皮细胞结构清晰,金颗粒呈团状,点灶状分布,特异性强,无非特异性散在金颗粒。结论:免疫电镜金标记技术应用中某些关键性技术环节的处理是获得理想免疫金染色切片的保证。

Abstract:Objective To explore the treatment of some key techniques of immunoelectron microscope colloid gold labelling technique. Methods Colloid gold labelling and immunoelectron microscope technique was used to locate and observe GST-Pi in colorectal adenocarcinoma,NGF (nerve growth factor) and bFGF (basic fibroblastgrowth factor) in neural tube of hamster embryo. Results The structure of colorectal mucosa cells and neural cells was clear. Colloid gold particles were distributed in spotty group. Conclusion Colloid gold labelling and immunoelectron microscope technique is a valid method. The treatment of some key techniques is the guarantee of getting good result.
