| 摘 要： | 目的 建立Bcl-2硫代反义核酸(G3139)OPC纯化方法。方法 将G3139从合成柱上解离并去保护，用OPC柱分离纯化化学合成的G3139。紫外分光光度计测OD值，20％PAGE鉴定其纯度。结果 经OPC柱纯化后G3139制备量可达mg水平，回收率为66％，电泳结果显示为一条亮带。结论 Bcl-2硫代反义核酸OPC纯化法是一种高效、快速、简便的方法。

| Abstract: | Objective\ To develop a method of OPC purification of antisense phosphorothioate oligodeoxynucleotide to Bcl 2.Methods\ After completion of the trityl on synthesis,G3139 was cleaved from the DNA synthesis column and deprotected,G3139 was purified by OPC column and its concentration was determinated by OD(A260),The purity of G3139 was analyzed by 20% PAGE method.Results\ The yields of purified G3139 could reached mg level and the productive rate being 66%,A single band was obseved from the result of 20% PAGE.Conclusion\ OPC colum provides a high effciency,quick and convenient method for the purification of antisense phosphorothioate Oligodeoxynucleotide to Bcl 2. |
