两种动脉粥样硬化兔模型制作方法的比较

目的通过对比单纯高脂饲养法与高脂饲养联合颈总动脉球囊损伤法建立动脉粥样硬化兔模型的试验研究,比较两种方法在制模时间,模型稳定性方面的差异.方法雌性新西兰兔20只,随机分成高脂饲养组（n=10）;高脂饲养＋颈动脉球囊拉伤组（n=10）,高脂饲养组给予高脂饲料喂养8周,颈动脉球囊拉伤组于喂养后第2周行颈动脉球囊拉伤术,术后继续喂养6周.所有试验兔在高脂喂养前后分别检测血脂水平,于第8周处死动物,取损伤段血管固定,切片,通过HE染色、弹力纤维染色后拍照,通过图像分析软件测量血管内膜厚度和泡沫细胞面积率的变化.结果高脂饲养8周形成明显的高脂血症;与单纯高脂饲养组相比,球囊损伤法颈动脉内膜明显增厚,泡沫细胞的阳性面积率高.结论与单纯高脂饲养法相比高脂饲养＋球囊损伤组试验兔8周即可形成明显的动脉粥样性血管病变,成膜时间快,泡沫细胞的阳性面积率高,模型稳定性好。

The Comparison between Two Methods of Making the Atherosclerotic Rabbit Model

Objective To compare the diffierence between high-fat feeding and high-fat diet combined with carotid artery balloon injury in making atherosclerotic rabbit model,to evaluate the two methods in modeling time and stability of the atherosclerotic rabbit model.Methods Twenty female New Zealand white rabbits were randomly divided into high-fat feeding group（n =10） and high-fat diet combined with balloon injury group（n=10）,all rabbits were fed with high-fat diet for 8 weeks,after 2 weeks we randomly selected 10 rabbit form high-fat diet group and injured right carotid artery with balloon,after balloon injury,those rabbits were still fed with high-fat diet for 6 weeks.We measured TG,LDL,HDL,CHO levels in rabbits＇serum before and afer high-fat diet feeding.At the 8th week we executed all rabbits,and got the right carotid artery.The samples were stained with HE to observe the positive rates of foam cells,samples were also stained by elastic fibers to calculate the thickness of medial area.Results The evident hyperlipidemia was formed in rabbits after high-fat diet feeding for 8 weeks.Compared with high-fat diet feeding group,the thickness of carotid artery intima was thicker and the positive rate of foam cells was more in high-fat diet combined with balloon injury group.Conclusion Compared with single high-fat diet feeding,the method of high-fat diet feeding combined with balloon injury can successfully make atherosclerotic rabbit model after 8 weeks,and can shorten the modeling time and improve the stability of atherosclerotic rabbit model
