舌鳞癌抗原提呈相关基因表达的研究

目的：分析舌鳞状细胞癌组织中抗原提呈相关基因的表达。方法：选取2008年12月～2009年12月在南京市口腔医院口腔颌面外科就诊的30例原发舌癌患者的肿瘤及癌旁组织标本,采用实时定量PCR检测TAP-1和Tapasin的mRNA相对表达量,同时检测正常黏膜角化上皮细胞及舌鳞癌细胞系中TAP-1和Tapasin的表达。结果：与癌旁组织相比,肿瘤标本中TAP-1和Tapasin的表达显著下降,并且其mRNA表达水平与肿瘤的病理分级有显著相关性（P=0.001）。与正常口腔黏膜上皮细胞相比,肿瘤细胞中TAP-1和Tapasin的表达亦呈明显降低。结论：舌鳞癌中TAP-1和Tapasin的表达明显降低,可能是导致肿瘤发生免疫逃逸的机制之一。

Study on Antigen Processing Machinery Genes in Tongue Squamous Cell Carcinoma.

Objectives： To investigate the expression level of antigen processing machinery（APM） genes in tongue squamous cell carcinoma（SCC）.Methods： 30 tongue SCC patients treated in Oral and Maxillofacial Surgery Department,Nanjing Stomatological Hospital,from December 2008 to December 2009 were analyzed.Realtime PCR was applied to detect the relative mRNA expression of TAP-1 and Tapasin in tumor samples and tumor cell lines.Results： Compared with the adjacent normal tissue samples,the relative mRNA expression of TAP-1 and Tapasin in cancerous tissue samples was downregulated.It had significant correlation with the pathological differentiation grade.The mRNA expression of TAP-1 and Tapasin was also downregulated in cancer cell lines,compared with KC cells.Conclusion： Expression of TAP-1 and Tapasin was downregulated.It could be one of possible mechanisms for immune escape for OSCC.
