转录因子NF-κB在内毒素休克中的作用

摘    要:目的探讨内毒素休克大鼠组织炎性介质的表达特征及其和核转录因子NFκB(nuclearfactorkappaB)的关系。方法应用免疫组织化学技术检测脂多糖(lippolysaccharice,LPS)内毒素休克大鼠肝肾组织转录因子NFκB、炎性介质ICAM1、VCAM1、iNOS的表达。结果LPS内毒素休克大鼠肝肾组织转录因子NFκBp65,炎性介质ICAM1、VCAM1、iNOS阳性细胞率高于正常对照组;炎性介质ICAM-1、VCAM-1、iNOS阳性细胞率与NF-κBp65阳性细胞率成正相关。用吡咯烷二硫氨基甲酸(pyrrolidinedithiocarbmate,PDTC)抑制转录因子NFκB的内毒素休克大鼠炎性介质ICAM1、VCAM1、iNOS阳性细胞率低于LPS组。结论核转录因子NFκB在LPS引起的大鼠内毒素休克炎性介质的表达中起调节作用。

Abstract:Objective To explore the role of transfection nuclear kappa B(NF-κB) in the pathogenesis of endotoxin shock. WT5”HZ]Methods The expressions of NF-κB,ICAM-1(intercellular adhesion molecule-1),VCAM-1 (vascular cell adhesion molecule-1) and iNOS (inducible nitric oxide synthase) in the kidney and liver tissue of SD rats were determined by immunohistochemistry.Results The expressional rates of NF-κB p65, ICAM-1,VCAM-1 and iNOS positive cells in the liver and kidney tissue of SD rats were increased in the endotoxin shock group than in normal control group, whereas the expressional rates of ICAM-1,VCAM-1 and iNOS positive cells in the liver and kidney tissue of SD rats were lower in LPS-PDTC (pyrrolidine dithiocarbmate) injection group than in LPS group.The expressional rate of NF-κB p65 cells was positively correlated with the expressional rate of ICAM-1,VCAM-1 and iNOS cells in the endotoxin shock group and LPS-PDTC injection group.Conclusion NF-κB may play a pivotal role in the pathogenesis of endotoxin shock in the regulation of the expression of inflammatory factors.
