清金益肺汤对痰热蕴肺型特发性肺纤维化大鼠发挥抗氧化作用的实验研究

摘    要:目的:通过检测痰热蕴肺型特发性肺纤维化大鼠应用清金益肺汤后血清超氧化物歧化酶(SOD)、还原型谷胱甘肽(GSH)的活性及光镜下大鼠肺组织的病理变化,探讨清金益肺汤对于痰热蕴肺型特发性肺纤维化病程中氧化/抗氧化失衡的调节作用。方法:清洁级健康雄性SD大鼠40只,随机分为5组:空白对照组(K组)、痰热蕴肺型特发性肺纤维化模型组(M组)、中药治疗组(Z组)、激素治疗组(J组)、中药加激素治疗组(H组)。采用气管内滴博来霉素的方法建立肺纤维化模型,造模结束后第二天滴加脂多糖建立痰热蕴肺证模型。光镜下观察各组大鼠肺组织的病理改变;化学荧光法检测各组大鼠腹主动脉血清SOD、GSH的活性。结果:M组、Z组、J组、H组在光镜下均有肺纤维化的特征性病理改变,但程度有所不同。与K组相比,M、Z、J、H组SOD、GSH的活性明显降低,有显著统计学意义(P<0.01)。与M组相比,Z、J、H组SOD、GSH活性显著升高(P<0.01);且H组明显高于Z组和J组。J组SOD活性明显低于Z组(P<0.01),而J组和Z组的GSH活性相较没有统计学意义(P>0.05)。结论:痰热蕴肺型特发性肺纤维化大鼠肺内存在氧化应激,清金益肺汤可明显增加肺内SOD、GSH的含量,使其在抗氧化机制中发挥重要作用,从而说明清金益肺汤可纠正痰热蕴肺型肺纤维化的氧化/抗氧化失衡。

A probe into antioxidant effect of Qingjinyifei decoction on rats sufferring from syndrome of phlegm - heat obstructing lung of idiopathic pulmonary fibrosis

Abstract:Objective:To explore the oxidant / antioxidant effect of Qingjinyifei decoction on rats sufferring from syndrome of phlegm - heat obstructing lung of idiopathic pulmonary fibrosis, by detecting the activity of SOD and GSH in serum and pathological changes of lung tissue under light microscope. Methods : Healthy male SD rats of clean grade were randomly divided into five groups : Blank control group ( K group) , syndrome of phlegm - heat obstructing lung of idiopathic pulmonary fibrosis model group (M group) ,traditional Chinese medicine therapy group( Z group) , hormone therapy group (J group) , traditional Chinese medicine plus hormone therapy group ( H group ). Establish the idiopathic pulmonary fibrosis models by dropping the bleomycin into trachea, and establish the phlegm - heat obstructing lung models by dropping the lipopolysaccharide into trachea the next day. Observe pathological changes of lung tissue under light microscope, and detect the activity of SOD and GSH in abdominal aorta serum in each group. Results:M group, Z group ,J group and H group had different degrees of characteristic pathological changes of pulmonary fibrosis under the light microscope. Compared with the K group, the activity of SOD and GSH decreased significantly in M, Z ,J and H group,which had statistical significance ( P 〈 0.01 ) ; the level of M group was lower than Z ,J and H group( P 〈 0. 01 ) ; H group was higher than Z and J group(P 〈0.01 ). The activity of SOD in J group was lower than Z group(P 〈0.01 ) ,and it had no significant difference in the activity of GSH between Z and J group (P 〉 0.05 ). Conclusion:There exists oxidative stress in rats sufferring from syndrome of phlegm - heat obstructing lung of idiopathic pulmonary fibrosis. Qingjinyifei decoction can obviously increase the content of SOD and GSH in lung, which plays an important part in mechanism of antioxidant. All of these prove that Qingjinyifei decoction can correct the oxidant / antioxidant imbala
