中药复方对Lewis肺癌移植鼠肿瘤微环境中吲哚胺2,3-双加氧酶表达的影响

摘    要:目的:研究中药复方对Lewis肺癌移植鼠肿瘤微环境中吲哚胺2,3-二氧化酶(indoleamine 2,3-dioxygenase,IDO)表达的影响。方法:选用近交系C57BL/6纯系小鼠64只,将Lewis肺癌细胞悬液接种于小鼠右腋皮下建立肺癌模型,接种后小鼠随机分为4组:对照组、化疗组、中药复方组和中药复方加化疗组,通过应用免疫组化法及免疫印迹(Western blot)方法等多种实验技术,分别检测Lewis肺癌移植鼠肿瘤及淋巴组织中IDO表达水平。结果:中药复方及化疗药物顺铂均可在不同程度上影响IDO表达水平。同时中药复方与化疗药物顺铂具有协同作用,两者在不同环节调控调节性T细胞的作用与单一治疗相比存在一定优势。结论:中药复方与化疗在消除肿瘤微环境的局部免疫耐受中具有协同作用。

Abstract:Objective:To study the influence of a traditional Chinese medicine (TCM) herbal compound on the expression of in- doleamine 2,3-dioxygenase(IDO) in the tumor microenvironment of transplanted Lewis lung cancer mice. Methods: A total of 64 inbred C57BL/6 mice were chosen, which were inoculated Lewis lung carcinoma cell suspension in their right axilla subcuta- neously. After that, they were divided randomly into 4 groups: control group, chemotherapy group, traditional Chinese medi- cine group and traditional Chinese medicine plus chemotherapy group. A variety of experimental techniques, such as immuno- histochemistry and Westerm blot method were used to detect IDO expression level in the tumor and lymphoid tissues of trans- planted Lewis lung cancer mice. Results: The traditional Chinese medicine and chemotherapy drug cisplatin could affect the IDO level in varying degrees. Traditional Chinese medicine had a synergistic effect with cisplatin, and there were therapeutic advan tages in regulating regulatory T cells. Conclusions.. The traditional Chinese medicine and chemotherapy have a synergistic effect in eliminating local immune tolerance in tumor microenvironment.
