碱性成纤维细胞生长因子对不同日龄大鼠癫(癎)持续状态后海马神经发生的影响

摘    要:目的了解不同年龄大鼠癫间疒持续状态后外源性碱性成纤维细胞生长因子(bFGF)对海马齿状回颗粒细胞层神经发生的影响。方法建立14天大鼠和60天大鼠戊四氮(PTZ)诱导癫疒间持续状态模型,生理盐水(NS)注射作为对照,皮下注射bFGF进行干预,分4组:NS组、NS bFGF组、PTZ组、PTZ bFGF组。采用B rdu标记新生细胞,免疫组化方法检测造模后大鼠海马齿状回神经发生情况,造模后第7、14天2个时间点观察海马齿状回B rdu阳性细胞数。结果造模后第7天和14天,PTZ组中各日龄组大鼠海马齿状回B rdu阳性细胞数明显高于各自对应的NS组(P<0.01),NS bFGF组各日龄大鼠B rdu阳性细胞数亦高于NS组(P<0.05),PTZ bFGF组B rdu免疫阳性细胞数较PTZ组亦有升高(14天日龄组P<0.05,60天日龄组P<0.01)。60天大鼠较14天大鼠升高幅度更为明显(P<0.01)。结论bFGF可增加癫疒间持续状态大鼠海马齿状回颗粒细胞发生,对60日龄大鼠比14日龄幼鼠更为明显。

Abstract:Objective To study the neurogenesis of hippocampal dentate gyrus granular cell layer in different-age rats with induced status epilepticus and its response to ectogenic basic fibroblast growth factor(bFGF).Methods Pentetrazole(PTZ) was used to form status epilepticus model on 14-day and 60-day old rats.The experiment was carried out in four groups,with the results analysed 7 days and 14 days after treatment.Brdu labeled new cells were counted to describe the neurogenesis in dentate gyrus.Results 7 days and 14 days after treatment,Brdu positive cells in the dentate gyrus were more numerous in the PTZ group and the NS bFGF group than in the NS group,more numerous in the PTZ bFGF group than in the PTZ group(P<0.01,P<0.05).The increment was more marked in the 60-day-old rats than in the 14-day-olds.Conclusion Basic fibroblast growth factor can promote neurogenesis in the hippocampus dentate gyrus granular cell layer in rats with status epilepticus,and the change is more marked in the elder rats than in the younger.
