Abstract:Black peanut skin pigment(PBSP)was extracted and purified from the black peanut skins which were regarded as a by-product of the peanut industry,and its related cell biological activities were preliminarily studied.First,the detection conditions,temperature and pH stability and optimum extraction conditions of BPSP were explored by UV-vis full-spectrum scanning technique and spectrophotometry.Results showed that BPSP has optimal characteristic absorption peak at pH2~3 and 520nm visible wavelength,and it is more stable in aqueous solution than in ethanol solution;also,BPSP aqueous showed the best stability when the temperature is not higher than 50℃at pH2.0;in addition,the optimal extraction conditions of BPSP were as follows:70℃,pH2.0 acidic aqueous solution extract for 15min;under this condition,the yield of BPSP was 7.95mg/g dry black peanut coat weigh.Further,BPSP anthocyanins were purified by ethyl acetate(EtOAc)partition combined with macroporous resin column chromatography technique,and the purity of total anthocyanins was increased nearly by 5-folds,from 5.2%(raw extraction)to 25.4%(after purification).Finally,purified BPSP was used to intervene the differentiated mature C2C12 myotubes.It was found that BPSP at the dose of 5μg/mL could significantly increase the glucose consumption of C2C12 myotube cells(P<0.05),moreover,within the concentration range of 1~5μg/mL,this promoting effect was dose-dependent.
