补肾安胎方对胚泡着床障碍小鼠子宫LIF及其受体表达的影响

摘    要:目的:观察补肾安胎方对胚泡着床障碍小鼠子宫白血病抑制因子(Leukaemia inhibitory factor,LIF)及其受体(Leukaemia inhibitory factor receptor,LIFR)表达的影响。方法:将雌鼠随机分为未孕组、正常组、模型组、中药组,每组40只,在妊娠4、5、6天(Pd4、5、6)均取妊娠子宫-80℃保存。采用免疫荧光双标和免疫印迹法分别观察LIF及其受体的表达。结果:免疫印迹显示正常组和中药组Pd4LIF蛋白为表达高峰,高于模型组(P<0.01),此后下降;模型组LIF蛋白表达高峰延迟至Pd5,高于正常组和中药组(P<0.01),此后下降。免疫荧光双标显示正常组和中药组LIF及其受体Pd4荧光较强,LIF及其受体表达基本同步;Pd4模型组LIF及LIFR均有较少表达,但LIFR表达更弱,其范围及强度不如LIF广泛,二者表达强度不完全同步;Pd5为模型组LIF及LIFR表达高峰,此后表达强度降低。结论:补肾安胎方可能通过促进LIF及其受体的同步表达来促进胚泡着床。

Abstract:Objective:To observe the effect of Bushenantai recipe on the expression of endometrial LIF and LIFR in the mouse with embryonic implantation dysfunction.Methods:Firstly, 40 Kunming mice post coition were randomly assigned to none pregnancy, normal, model and traditional Chinese medicine groups. On Pd 4, 5, 6, the uteruses of mice in each group were obtained at -80℃. And then, Western blot and double-labelling technique of immunofluorescence were used to detect and analyze the expression of LIF and LIFR protein.Results:Firstly, the time of high expression of LIF and LIFR protein was Pd4 in normal group and traditional Chinese medicine group were higher than those of model group (P<0.01 ), and then gradually depressed by Western blot. But the time of the model group delayed to Pd5 was higher than that of normal group and traditional Chinese medicine group (P<0.01) and then depressed gradually. Secondly, the fluorescence of LIF and LIFR of Pd4 were more powerful than other day in normal group and traditional Chinese medicine group, the expression of LIF and LIFR was in-synchronism by double-labelling technique of immunofluorescence. But the expression of LIF and LIFR of model group was few and LIFR was fewer, the expression was asynchronous between them. The powerful fluorescence of LIF and LIFR of model group was on Pd5, and then depressed.Conclusion:Improving the in-synchronism expression of LIF and LIFR in endometrium may be one of the mechanisms for Bushenantai recipe to improve embryo implantation.
