对环境扫描电子显微镜(ESEM)的认识

摘    要:环境扫描电子显微镜(ESEM)中所指环境并非真正意义上的大气环境,与传统SEM样品室高达10∧-3~10∧-6Torr的真空度相比,ESEM样品室的真空度很低,从1Torr~20Torr,非常接近大气环境,但不等同于平均760Torr的大气环境;环境方式下最适家观察的生物样品是那些表面有角质层覆盖的样品和含水量很低的样品,比如植物的叶片、动物中的昆虫、作物的籽粒等,这类样品易于保持新鲜度;环境方式下对样品的观察和图像的记录等操作应快完成,这样可减少样品内水分蒸发而使样品变形降至最低。

Abstract:So called environment in environmental electron microscope does not really mean atmosphere. Compared with conventional scanning electron microscope, the vacuum in its chamber is rather low ranging from 1~20 Torr, being not equal to the atmosphere. The most appropriate biological samples observed in environmental manner are those which contain a small amount of water and could keep fresh, such as leaves, insects and grains. To avoid the sample deformation resulted from the water evaporing, it is important to complete the observation and recording as quick as possible.
