海马干细胞移植对癫癎鼠脑电影响的初步研究

摘    要:目的研究神经干细胞移植对癫癎鼠脑电的影响,为神经干细胞移植治疗癫癎提供理论依据.方法分离、培养新生鼠海马干细胞,将其移植至海人酸(KA)所致癫癎模型鼠的右侧海马,比较移植组及未移植组大鼠在移植前和移植后1周、4周、8周及24周海马及杏仁核的脑电变化.结果海马干细胞移植可减少癫癎动物脑电的痫性发放,并降低癫癎波幅约50%.结论神经干细胞移植对于KA诱发癫癎鼠的癫癎具有一定的抑制作用,但其具体作用机制还有待于进一步的研究.

Abstract:Objective To investigate the effects of neural stem cells transplantation on electroencephalogram in epileptic rats, and provide some theoretical bases on the therapy of epilepsy with neural stem cells transplantation. Methods Neonatal rat hippocampal stem cells were isolated and cultured in the culture medium without serum, and then were transplanted into the right hippocampus of the kainite-induced epileptic rats. The electroencephalogram of right hippocampus and the amygdala nucleus were compared between the transplanted group and the non-transplanted group after 1, 4, 8 and 24 weeks. Results Hippocampal stem cells transplantation can decrease the frequency of discharge of the epileptic rats, and reduce the amplitude of discharge to half its previous level. Conclusions Neural stem cells transplantation has some inhibitory effect on the epilepsy of kainite-induced epileptic rats, and its principle should be researched more.
