抗癌药1,2:5,6-二去水卫矛醇与长春新碱联合用药的实验研究

摘    要:去水卫矛醇(DAG)与长春新碱(VCR)联合使用,对小鼠淋巴细胞白血病L_(1210)有协同疗效,疗效强度决定于给药方案和剂量。先给DAG,24 h后给VCR,协同疗效最高。DAG剂量是影响疗效的主要因素,最佳剂量为3.0 mg/kg。琼脂扩散合试验亦证明二药有方案依赖性协同作用。二药合用对小鼠骨髓干细胞仅有微弱的协同杀灭作用。

Abstract:The combined effect of 1,2:5,6-dianhydrogalactitol (DAG) and vincristine (VCR) on L_(1210) leukemia was studied. The results obtaned in vivo suggest that simultaneous administration of DAG and VCR, or administration of these two agents within a 24 h interval, independent of order, produced a synergistic effect. The degree of synergism was dependent on dose and schedule. When DAG was given before VCR, the effect was more than the other two schedules of combination, Studies using different doses of DAG and VCR with an optimum schedule indicated that the degree of synergism was mainly dependent on the dose of DAG. When a dose of 3.0 mg/kg of DAG was given before 1.3 mg/kg of VCR within a 24 h interval, the survival time of the L_(1210)-bearing mice was four times as long as additive, but the cell killing effect of the combination on the bone marrow stem cells was only increased by 12% over the additive. The schedule-dependent synergism for L_(1210) cells was also studied by the agar-diffusion-chamber assay.
