中国仓鼠肺细胞胞质分裂阻滞法微核试验灵敏性初探

摘    要:作者测试了断裂剂与非整倍剂对细胞松驰素B诱发中国仓鼠肺细胞(CHL)双核细胞率的影响;对诱变剂诱发常规法单核细胞,胞质分裂阻滞法单、双核细胞微核率进行比较研究。结果显示:丝裂霉素C对细胞松驰素B诱发双核细胞率有降低作用,而秋水仙素有增加作用;诱变剂诱发的双核细胞微核率并不显著高于常规法核细胞微核率。该结果胞质分裂阻滞法微核试验不能显著提高微核试验的灵敏性。作者并分析了其影响因素。

Abstract:Studies were performed to determine the cytochalasin B induced binucleated cell percentage influenced by clastogens and aneuploidgens and to compare the efficacy of cytokinesis blocked binucleated cells for scoring micronuclei with that of the conventional mononucleated method following the treatment with mitomycin C, methyl methanesulforate, colchicin and chloral hydrate. The results showed that mitomycin C decreased the binucleated cell percentage induced by cytochalasin B, whereas colchicin increased the frequencies of binucleated cells. The frequencies of micronuclei in binucleated cells were not significantly higher than those in the conventional mononucleated cells. The results suggest that cytokinesis blocked method is not more sensitive than the conventional method for scoring micronuclei. The factors that may influence the cytokinesis blocked micronucleus assay have been discussed.
