MMP-2,9在羊膜移植治疗小鼠单疱病毒性角膜炎中的表达

摘    要:目的:观察保存人羊膜移植(AMT)对单疱病毒性角膜基质炎(HSK)中基质金属蛋白酶(MMPs)在小鼠角膜中表达的影响,从而探讨羊膜移植对MMP-2,9的调节作用及对HSK治疗的潜在作用。方法:雌性BALB/c小鼠眼角膜接种Ⅰ型单纯疱疹病毒(HSV-1,SM44毒株)以诱发单纯疱疹性角膜炎。选取右眼感染2 d后出现典型树枝状上皮病变的小鼠40只,随机分为实验组和对照组,实验组小鼠右眼角膜行AMT,对照组仅缝合睑裂不行AMT。术后第0,2,7,14天处死动物取出角膜,应用Western blot方法对结果进行分析,增强化学发光法(ECL)检测小鼠角膜中MMP-2,9的表达。结果:在对照组,角膜中MMP-2,9在第2天表达增加,第7天表达暂时减弱,第14天时达高峰。实验组羊膜移植2 d后角膜炎症反应明显消退且上皮缺损较快愈合,MMP-2,9阳性表达减弱,各时间点MMP-2,9的表达均低于对照组,其差异均有显著性意义(P<0.01)。结论:羊膜移植术可通过抑制角膜细胞及浸润的炎症细胞产生的MMP-2,9,促进快速上皮化并减轻角膜基质炎症反应,抑制角膜新生血管化和溃疡的形成。

Abstract:Objective: To study the expression of matrix metalloproteinases(MMPs) in the corneas from mice with Herpes stromal keratitis(HSK) treated with amniotic membrane transplantation(AMT) and to investigate the possible regulative mechanism of MMP-2,9 after AMT and the potential effect in the treatment of the HSK.Methods: Forty BALB/c mice were infected with herpes simplex virus type-1(HSV-1)(SM44 strain) to induce HSK.After two days,the mice were divided equally and randomly into the AMT group and the control group,in which each mouse was only treated with tarsorrhaphy.After different time (0,2,7 and 14 d),the expression of MMP-2 and MMP-9 in corneas was measured by the Western blot technique.Results: In the control group,the expression of MMP-2 and MMP-9 in corneas was increased at 2 d,decreased at 7 d and peaked at 14 d.In the AMT group,corneas inflammation and inflammatory cells infiltration were markedly improved after AMT.The expression of MMP-2 and MMP-9 in the AMT group was lower than in the control group.At the same point, the expression of MMP-2 and MMP-9 were less in the AMT group with significant differences(all P<0.01).Conclusion: These observations suggest that the improvement of stromal inflammation and reduced corneal neovascularization after AMT may result from the reduced expression of MMP-2 and MMP-9.
