抑制TRIM24基因表达对乳腺癌MCF-7细胞增殖、凋亡、侵袭和迁移的抑制作用

摘    要:目的:探讨抑制TRIM24基因表达对乳腺癌MCF-7细胞增殖、凋亡、侵袭和迁移的影响,阐明TRIM24基因在乳腺癌发生发展中的作用。方法:以脂质体法转染TRIM24 siRNA和阴性对照NC-siRNA至MCF-7细胞作为si-TRIM24组和NC-siRNA组,以未转染的MCF-7细胞作为对照组。qRT-PCR和Western blotting法检测转染48 h后各组MCF-7细胞中TRIM24 mRNA和蛋白表达水平,MTT法检测各组MCF-7细胞增殖能力,流式细胞术检测各组MCF-7细胞凋亡率,Transwell实验检测各组MCF-7细胞侵袭和迁移数。结果:与对照组和NC-siRNA组比较,si-TRIM24组MCF-7细胞中TRIM24 mRNA和蛋白表达水平明显降低(P<0.05),MCF-7细胞增殖能力降低(P<0.05),MCF-7细胞凋亡率明显升高(P<0.05),MCF-7细胞侵袭和迁移数明显减少(P<0.05)。结论:抑制TRIM24基因表达能够明显抑制乳腺癌MCF-7细胞的增殖、侵袭和迁移,且能够诱导细胞凋亡。

Abstract:Objective: To investigate the effect of inhibiting TRIM24 gene expression on the proliferation, apoptosis, invasion and migration of breast cancer MCF-7 cells, and to elucidate the role of TRIM24 gene in the occurrence and development of breast cancer. Methods: The TRIM24 siRNA and negative control NC-siRNA were transfected into the MCF-7 cells by liposome method, and used as si-TRIM24 group and NC-siRNA group, respectively. The untransfected MCF-7 cells were used as control group. qRT-PCR and Western blotting methods were used to detect the expression levels of TRIM24 mRNA and protein in the MCF-7 cells in various groups after transfected for 48 h,MTT assay was performed to detect the proliferation activities of the MCF-7 cells in various groups,flow cytometry was used to determine the apoptotic rates of the MCF-7 cells in various groups,and Transwell assay was used to measure the number of invasion and migration cells in various groups. Results: Compared with control group and NC-siRNA group, the expression levels of and protein in the MCF-7 cells were significantly decreased (P<0.05), and the proliferation activity of the MCF-7 cells was decreased (P<0.05), the apoptotic rate of the MCF-7 cells was significantly increased (P<0.05), and the number of invasion and migration MCF-7 cells was significantly decreased (P<0.05). Conclusion: Inhibition of the expression of TRIM24 gene can significantly inhibit the proliferation, invasion and migration of breast cancer MCF-7 cells, and induce the apoptosis.
