加工后亳州丹皮对大鼠心肌缺血再灌注损伤的保护作用研究

摘    要:目的探讨加工后亳州丹皮对大鼠心肌缺血再灌注损伤的保护作用及其机制。方法结扎大鼠左冠状动脉前降支30 min,再灌注60 min的方法复制大鼠心肌缺血再灌注损伤模型。用药前后测定大鼠血清中超氧化物歧化酶(SOD)、丙二醛(MDA)、磷酸肌酸激酶(CK)、乳酸脱氢酶(LDH)、谷胱甘肽(GSH)、一氧化氮(NO)的含量。HE染色观察大鼠心肌组织病理形态学变化,TUNEL法测定心肌细胞的凋亡。结果与模型组相比,加工后亳州丹皮组能明显降低血清中MDA,CK,LDH的含量,提高SOD,GSH,NO的含量;并能有效降低心肌细胞凋亡率。结论加工后亳州丹皮对大鼠心肌缺血再灌注损伤具有保护作用,其机制可能与清除氧自由基,增加NO的含量,抑制心肌细胞凋亡,减少心肌酶的漏出有关。

Abstract:Objective To investigate the protective effects and its mechanism of post-processed Bozhou Cortex Moutan on myocardial ischemia reperfusion injury rat.Methods The myocardial ischemia reperfusion injury model was established by a 30min occlusion of ananterolateral branch of the coronary artery,followed by a 60min reperfusion.The serum levels of SOD,MDA,CK,LDH,GSH,NO were determined to evaluate the effect of post-processed Bozhou Cortex Moutan on myocardial ischemia reperfusion injury in rat.The myocardial cell apoptosis was determined by TUNEL method.Results Compared with model group,post-processed Bozhou Cortex Moutan could significantly decrease levels of serum MDA,CK,LDH,improve the content of SOD,GSH and NO,and effectively reduce the myocardial cell apoptosis rate.Conclusion Post-processed Bozhou Cortex Moutan exerts protective effects on myocardial ischemia reperfusion injury.The mechanisms might be related to scavenging oxygen free radicals,increasing the content of NO,inhibiting myocardial cell apoptosis,and reducing the myocardial enzymes leaks out.
