银杏酚C171与顺铂联用逆转A549/DDP细胞对顺铂的耐药性

摘    要:探讨银杏酚C171对顺铂耐药人肺腺癌A549/DDP细胞株耐药及凋亡的影响。方法: 体外培养A549及A549/DDP细胞,MTT法检测A549/DDP细胞的耐药倍数,确定顺铂作用浓度;此浓度顺铂与不同浓度银杏酚C171(0, 10,20,40 mg/L)联合应用,分别通过Transwell法、免疫印记及流式细胞术检测A549/DDP细胞迁移、侵袭,Nrf2/Keap1信号通路相关蛋白的表达及细胞凋亡情况。结果: MTT结果显示,与A549细胞相比,A549/DDP细胞具有5.8倍耐药性,确定顺铂作用浓度为4 mg/L;与对照组相比,Transwell实验显示,各浓度银杏酚C171与顺铂联用均能明显抑制A549/DDP细胞的迁移和侵袭(P<0.05);免疫印迹结果显示,各浓度银杏酚C171与顺铂联用均能抑制A549/DDP细胞Nrf2、Mrp1及抗凋亡蛋白Bcl2的表达(P<0.05),且促进凋亡蛋白Bax及Keap1蛋白的表达(P<0.01);流式细胞术结果显示,各浓度银杏酚C171与顺铂联用均能致A549/DDP细胞凋亡率显著升高(P<0.05)。 结论: 银杏酚C171与顺铂联用可逆转体外A549/DDP细胞对顺铂的耐药性,并促进细胞凋亡。

Abstract:Objective: To explore the effect of ginkgol C171 on the antitumor drug resistance and apoptosis of cisplatin resistant lung adenocarcinoma (A549/DDP) cells. Methods: A549 and A549/DDP cells were cultured in vitro, and MTT assay was used to detect the drug resistance of A549/DDP cells and the optimal concentration of cisplatin. After treatment with cisplatin of the above optimal concentration and various concentrations of ginkgol C171 (0,10, 20, 40 mg/L), the migration, invasion, resistance related proteins and apoptosis rate of A549/DDP cells were detected by Transwell assay, Western blotting and flow cytometry, respectively. Results: MTT result showed the resistance of A549/DDP cells was 5.8 times vs A549 cells, and the optimal concentration of cisplatin was 4 mg/L. Compared with the control group, Transwell assay demonstrated that the migration and invasion of A549/DDP cells were significantly inhibited in ginkgol C171 at different concentrations and cisplatin combination groups(P<0.05). Western blotting showed that the expressions of Nrf2, Mrp1 and anti apoptotic protein Bcl2 were inhibited(P<0.05), while apoptotic protein Bax and Keap1 protein were increased(P<0.01) when A549/DDP cells were co treated with different concentrations of ginkgol C171 and cisplatin. Cytometry demonstrated that the apoptosis rates of A549/DDP cells were increased significantly by different concentrations of ginkgol C171 and cisplatin co treatment groups(P<0.05). Conclusion: Ginkgol C171 could reverse the resistance of A549/DDP cells to cisplatin in vitro,and promote apoptosis.
