免疫荧光法检测生殖支原体抗原的研究及临床意义

摘    要:目的研究间接免疫荧光技术(IFA)检测生殖支原体(Mg)抗原的敏感性及特异性,并探讨其临床价值。方法 以PCR为对照,采用Mg单克隆抗体作一抗行间接免疫荧光法检测生殖支原体抗原,对标准株及82份临床标本进行检测,比较其检出率和符合率。结果82例标本中IFA检测Mg抗原的敏感度为66.67%,特异度为100%,两种方法Mg检出率IFA为7.33%,PCR为10.95%,两者差异无显著性(P>0.05)。结论IFA检测法具有简便快速、特异性好和直观性强等优点。

Abstract:Objective To establish indirect immunofluorescence assay(IFA) method for mycoplasm genitalium(Mg) antigen detection using monoclonal antibodies specific to Mg and evaluate its value in clinical application. Methods Standard Mg strains and 80 clinical specimens were enrolled in this study. Monoclonal antibody specific to Mg was adopted to detect Mg antigen with IFA in these samples. PCR detection was conducted parallelly as control. Positive rate and coincidence rate between IFA method and PCR detection was analyzed too. Results Sensitivity of IFA for Mg antigen detection was 66. 67% and specificity was 100% . In 82 clinical specimens examined 6 positive in IFA and 9 positive in PCR detection. No significant difference of the positive rate was found between two methods(7.33% and 10.95% ). Conclusion The sensitivity and specificity of IFA method in Mg antigen detection are very good. Together with characters including faster to get result and easier to handle,and etc, IFA method is quite valuable for clinical Mg antigen detection.
